摘 要:目的探讨体外分离培养人绒毛膜来源间充质干细胞的方法,评价诱导的间充质干细胞分化为血管内皮细胞及体外成血管的可行性。方法从人胎盘绒毛膜组织中分离间充质干细胞及培养,采用流式细胞术检测细胞表面抗原表达;培养第2代细胞进行血管内皮细胞分化诱导,2周后采用免疫荧光染色法鉴定Matrigel基质胶中CD31和vWF的表达。结果从人胎盘绒毛膜组织中分离培养的细胞表达CD73、CD90和CD105,不表达CD34和HLA-DR。人绒毛膜间充质干细胞向血管内皮细胞定向诱导2周后,形态由长梭状逐步变为圆形、铺路石状,并在Matrigel基质胶上培养12 h后有管腔样结构形成,免疫荧光染色显示管腔样结构的细胞表达血管内皮细胞表面标记物CD31和vWF因子。结论胎盘绒毛膜可用于分离培养间充质干细胞,培养的细胞具有诱导分化为血管内皮细胞的潜能。Objective To explore the methods to isolate and culture human chorion-derived mesenchymal stem cells(MSCs),and to evaluate the feasibility of differentiation of MSCs into vascular endothelial cells in vitro.Methods MSCs were isolated from human placental chorionic tissue and cultured in vitro.Cell surface antigen expression was detected by flow cytometry.The cells in second passage were used for induction of endothelial differentiation.After 2 weeks,the expression of CD31 and vWF in Matrigel was identified by immunofluorescence staining.Results The human chorion-derived MSCs expressed CD73,CD90 and CD105,but did not express CD34 and HLA-DR.After MSCs were induced for 2 weeks,the cells gradually changed from fusiform shape to circle or paving stone shape.Furthermore,a lumen-like structure was formed after 12 hours of culture on Matrigel.Immunofluorescence staining showed that cells in the lumen-like structure expressed vascular endothelial cell surface markers CD31 and vWF.Conclusion Placental chorion can be used to separate MSCs,and the cultured cells have the potential to differentiate into vascular endothelial cells.
