摘 要:目的:探讨骨碎补总黄酮对悬尾实验性骨质疏松模型大鼠骨密度(BMD)及骨形成蛋白(BMP-2)mRNA的影响。方法:将48只6个月龄SD大鼠随机分为中药组、西药组、阴性对照组和空白对照组,每组12只。中药组予悬尾、骨碎补总黄酮灌胃(6.75 mg/mL);西药组予悬尾、阿仑膦酸钠灌胃(0.09 mg/mL);阴性对照组予悬尾、饮用水灌胃(1 mL/100g);空白对照组自由活动,持续灌胃28 d。采用双能X线检测大鼠的BMD,采用RT-PCR法检测骨组织BMP-2 mRNA表达情况。结果:中药组、西药组、阴性对照组大鼠骨密度及BMP-2 mRNA表达均低于空白对照组,差异均有统计学意义(P<0.05);中药组和西药组大鼠骨密度及BMP-2 mRNA表达高于阴性对照组,差异均有统计学意义(P<0.05);中药组与西药组比较,差异无统计学意义(P>0.05)。结论:骨碎补总黄酮能上调悬尾实验性骨质疏松模型大鼠BMP-2m RNA的表达,提高大鼠骨密度。Objective:To investigate the effects of total flavonoids of Drynaria on bone density(BMD)and bone morphogenetic protein(BMP-2)mRNA of rats with suspended tail osteoporosis.Methods:A total of 48 SD rats aged 6 months were randomly divided into TCM group,western medicine group,negative control group and blank control group,12 rats in each group.The TCM group were treated with suspended tail and Drynaria total flavonoids by gavage,6.75 mg/mL.Western medicine group was given tail suspension and sodium alendronate by gavage,0.09 mg/mL;The negative control group was given tail suspension and drinking water,1 mL/100 g.The blank control group was given free exercise for 28 days.BMD of rats was detected by dual-energy X-ray,and BMP-2 mRNA expression in bone tissue was detected by RT-PCR.Results:The TCM group,western medicine group,negative control group showed lower BMD and BMP-2 mRNA expression than the blank control group,and the differences were statistically significant(P<0.05).The TCM group and western medicine group showed lower BMD and BMP-2 mRNA expression than the negative control group,and the differences were statistically significant(P<0.05).There was no significant difference between traditional Chinese medicine group and western medicine group(P>0.05).Conclusion:The Drynaria total flavonoids can up-regulate the expression of bone morphogenetic protein(BMP-2)mRNA in the rat model of suspended tail osteoporosis,and improve the bone density of the rat.
