摘 要:目的探讨半胱胺和胱氨酸对卵母细胞体外成熟(IVM)和胚胎发育潜能的影响。方法收集超促排卵周期中未成熟卵母细胞584枚,随机分为添加组(IVM培养液中添加半胱胺和胱氨酸)和不添加组(单纯使用IVM培养液),在卵母细胞培养成熟后行显微授精,观察两组的成熟率、受精率、卵裂率、第3天(D3)优胚率和优质囊胚形成率。结果无论MⅠ期还是GV期卵母细胞,IVM培养液中半胱胺和胱氨酸的添加均能显著提高D3优胚率和优质囊胚形成率(P<0.05),但成熟率、受精率和卵裂率两组间比较差异无统计学意义(P>0.05)。结论IVM培养液中半胱胺和胱氨酸的添加能显著提高人未成熟卵母细胞的胚胎发育潜能,但其机理仍有待于进一步研究。Objective:To explore the effects of cysteamine and cystine on in vitro maturation(IVM)of oocytes and embryonic development.Methods:A total of 584 immature oocytes were collected from controlled ovarian stimulation cycles.They were randomly divided into two groups:adding group(addition of cysteamine and cystine in IVM medium)and non-adding group(only IVM medium).The matured oocytes were used for ICSI.The rates of maturation,normal fertilization,cleavage,high-quality embryo rate on Day 3,and high quality blastocyst formation rate were compared between the two groups.Results:Addition of cysteamine and cystine in the medium of IVM significantly increased the rate of high-quality embryo on Day 3 and the rate of blastocyst formation(P<0.05),but there were no significant difference in maturation rate,fertilization rate and cleavage rate between the two groups(P>0.05).Conclusions:Addition of cysteamine and cystine in the medium of IVM can significantly improve the embryonic development potential of human immature oocytes.Its mechanism is still to be further studied.
