摘　　要：为了研究豹蛙抗瘤酶(ONC)与维生素C(VC)对肿瘤细胞的协同促凋亡作用,对大鼠肝癌RH-35细胞进行单药组和联合用药组处理。采用磺酰罗丹明B法测定细胞增殖抑制率,Hoechst染色检测细胞核变化。采用联合指数和等效线图解法评价联合用药效果,流式细胞法检测细胞的凋亡情况。试验结果表明:ONC单药组和VC单药组对RH-35细胞增殖的抑制作用均显著(P<0.01),联合用药组的促凋亡作用优于单药组(P<0.001)。VC协同ONC促进了RH-35细胞凋亡。In order to study synergistic apoptosis-promoting effect of onconase(ONC)and vitamin C(VC)on cancer cell,the rat hepatoma RH-35 cells were treated by single drug group and combination drug group.The inhibitory rate of cell proliferation was determined by sulforhodamine B method,and the cell nucleus changes were detected by Hoechst staining.The combined drug effects were evaluated by combination index and isobologram,and the cell apoptosis conditions were measured by flow cytometry.The test results show that the inhibiting effect of ONC single drug group and VC single drug group on RH-35 cells proliferation both are significantly(P<0.01),and the apoptosis-promoting effect of combination drug group is better than single drug group(P<0.001).VC and ONC synergistically promote the apoptosis of RH-35 cells.
