摘 要:目的观察黄连素对妊娠期糖尿病胎盘滋养层细胞AMPK/GLUT4信号通路和葡萄糖摄取能力的影响。方法以32例妊娠期糖尿病患者为研究对象,35例健康孕产妇为对照组,提取胎盘滋养层细胞,体外培养24 h,观察细胞24 h糖消耗量,测定腺苷酸活化蛋白激酶(AMPK)、磷酸化AMPK(P-AMPK)、葡萄糖转运体4(GLUT4)蛋白表达水平。同时设梯度浓度干预实验,观察不同浓度黄连素干预对各个指标的影响。结果妊娠期糖尿病患者胎盘滋养层细胞的24 h糖消耗量明显少于对照组,P-AMPK/AMPK、GLUT4表达低于对照组,差异有统计学意义(P<0.05)。黄连素干预后,其24 h糖消耗量有所增加,P-AMPK/AMPK、GLUT4表达有所升高,且随着干预浓度的递增变化程度依次增大,差异均有统计学意义(P<0.05)。结论黄连素可以促进AMPK磷酸化,从而激活AMPK/GLUT4信号通路,促进细胞对葡萄糖的摄取能力,可能为其改善妊娠期糖尿病胰岛素抵抗的重要机制之一。Objective To observe the effect of berberine on AMPK/GLUT4 signal pathway of placental trophoblastic cells of gestational diabetes mellitus. Methods Totally 32 pregnant woman(the pathological objects) and 35 healthy pregnant women(the controls) were selected, which placental trophoblastic cells were extracted and cultured. 24 h glucose consumption of the cultured cells were compared between groups, and the adenosine activated protein kinase(AMPK), phosphorylated AMPK(P-AMPK), glucose transporter 4(GLUT4) protein expression levels were detected by Western blot(WB) assay. Furthermore, a gradient concentration interference experiment was designed to observe the influence of berberine on each index. Results The 24 h glucose consumption of placental trophoblastic cells of gestational diabetes mellitus was less than that of the controls, the expressions of P-AMPK/AMPK and GLUT4 protein were higher than those of the controls, and the difference between groups had statistical significance(P<0.05). After berberine interferes, the 24 h glucose consumption, P-AMPK/AMPK and GLUT4 protein expressions of placental trophoblastic cells of gestational diabetes mellitus all increased, and the difference between the groups had statistical significance(P<0.05). Conclusion Berberine can promote AMPK phosphorylation and activate AMPK/GLUT4 signal pathway, so as to promote cell glucose uptake ability, which may be one of its important mechanisms to relieve insulin resistance in pregnancy.
