摘 要:目的探讨Toll样受体5(TLR5)基因单核苷酸多态性(single nucleotide polymorphism,SNP)rs2072493位点与幽门螺杆菌(Helicobacter pylori,H.pylori)感染的关系。方法选取包头地区健康体检者683名的血液样本,采用酶联免疫吸附测定法(enzymelinked immunosorbent assay,ELISA)检测所有样本中H.pylori的感染情况,根据抗体滴度将样本分为H.pylori阴性组(n=305)和H.pylori阳性组(n=378),采用聚合酶链式反应-限制性片段长度多态性法(polymerase chain reaction-restriction fragment length polymorphism,PCR-RFLP)对TLR5 rs2072493位点进行基因分型,采用非条件性Logistic回归分析在共显性、显性、隐性三种遗传模式下rs2072493与H.pylori感染的关系。结果在共显性遗传模式下,TLR5基因SNP rs2072493与H.pylori的感染关联,与AA基因型携带者相比,携带AG基因型者可使H.pylori感染的风险降低(OR=0.576,95%CI:0.358~0.927)。结论TLR5 rs2072493携带AG基因型者H.pylori感染风险降低。Objective To investigate the relationship between single nucleotide polymorphism(SNP)rs2072493 locus of Toll-like receptor-5(TLR5)gene and Helicobacter pylori(H.pylori)infection.Methods A total of 683 blood samples of physical examinees in Baotou area were selected.The infection of H.pylori in all samples was detected by enzyme-linked immunosorbent assay(ELISA).The samples were divided into H.pylori negative group(n=305)and H.pylori positive group(n=378)according to the titer of antibody.Polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP)was used to genotype the TLR5 rs2072493 locus.The relationships between rs2072493 and H.pylori infection in co-dominant,dominant and recessive genetic models were analyzed by unconditional Logistic regression.Results In co-dominant genetic model,SNP rs2072493 of TLR5 gene was associated with H.pylori infection.Compared with AA genotype,AG genotype could reduce the risk of H.pylori infection(OR=0.576,95%CI:0.358-0.927).Conclusion The AG genotype of TLR5 rs2072493 can reduce the risk of H.pylori infection.
