单链抗体在毕赤酵母中表达的研究进展

摘　　要：抗体是现代医学中一种十分重要的工具,被广泛用于生物学实验室研究、疾病诊断和临床治疗等。单链抗体是第三代抗体的一种,具有相对分子质量小、组织渗透性好、特异性强、免疫原性低及半衰期短等特点。目前,单链抗体在医学方面主要用于抗肿瘤、抗病原生物、自身免疫疾病研究、心血管疾病研究以及抗辐射损伤等;在食品安全方面主要用于食品毒素检测、农兽药物残留检测以及食品中重金属污染检测等。因而,单链抗体的获得与制备至关重要。本文主要综述了单链抗体的发展及应用、单链抗体在毕赤酵母中的重组表达及提高单链抗体在毕赤酵母中表达量可采取的策略。Antibody is a very important tool in modern medicine and is widely used in biological laboratory research,disease diagnosis and clinical treatment.The single-chain variable fragment(scFv)is one of the third generation antibodies,which has the characteristics of small molecular weight,good tissue permeability,high specificity,low immunogenicity and short half-life.At present,scFv are mainly used in medicine for anti-tumor,antipathogenic organisms,autoimmune diseases research,cardiovascular disease research and anti-radiation damage;in food safety,they are mainly used for food toxin detection and agricultural and animal drug residues,and detection of heavy metal pollution in food.Thus,the acquisition and preparation of scFv is critical.This article focused on the development and application of scFv,recombinant expression of scFv in Pichia pastoris,and strategies for increasing the expression of scFv in P.pastoris.
