摘  要:目的探讨硒对同型半胱氨酸(Hcy)诱导的内皮细胞功能紊乱的影响。方法18只雄性SD大鼠随机均分为三组:H组灌注左旋蛋氨酸1 g·kg^-1·d^-1,连续4周;S组在灌注左旋蛋氨酸前1周,灌注硒1 g·kg^-1·d^-1;对照组(C组)灌注蒸馏水。检测三组大鼠血浆一氧化氮和血管性血友病因子(vWF)含量,采用胸主动脉环松弛实验观察内皮依赖性松弛度。培养人脐静脉内皮细胞(HUVECs),用Hcy及不同浓度硒处理HUVECs,检测细胞活力、迁移及凋亡的变化。结果与C组比较,H组血浆一氧化氮含量减少,vWF含量增加(P<0.05);与H组比较,S组一氧化氮含量增加,vWF含量减少(P<0.05)。加入乙酰胆碱或一氧化氮合成酶活性剂A23187后,H组胸主动脉环松弛度较C组降低(P<0.05),S组松弛度较H组升高(P<0.05)。用Hcy处理HUVECs后,细胞活力和细胞迁移能力降低,细胞凋亡增加,Caspase-3活性升高,剪切型Caspase-3、Bax表达增加,Bcl-2表达减少(P<0.05);用不同浓度硒预处理HUVECs后,细胞活力和细胞迁移能力升高,细胞凋亡减少,Caspase-3活性降低,剪切型Caspase-3、Bax表达减少,Bcl-2表达增加(P<0.05)。结论硒能够拮抗Hcy诱导的内皮细胞功能紊乱。Objective To investigate the effect of selenium on homocyste-induced endothelial cell dysfunction.Methods Eighteen SD male rats were divided into three groups with 6 rats each.The rats in group H were perfused with L-methionine 1 g·kg^-1·d^-1 for 4 weeks,those in group S were perfused with selenium 1 g·kg^-1·d^-1 one week before L-methionine perfusion and those in group C were perfused with distilled water.Plasma levels of nitric oxide and von willebrand factor(vWF)were detected in three groups.Endothelial dependent relaxation was observed by relaxation test of thoracic aortic ring.Human umbilical vein endothelial cells(HUVECs)were cultured and treated with homocysteine and different concentrations of selenium.The cell viability,migration and apoptosis were evaluated.Results Compared to group C,plasma level of nitric oxide was lower and vWF was higher in group H(P<0.05).Plasma level of nitric oxide was higher and vWF was lower in group S than those in group H(P<0.05).The relaxation of thoracic aortic ring was lower in group H than that in group C after acetylcholine or A23187 was added(P<0.05),which was higher in group S than that in group H(P<0.05).The cell viability and migration were lower,cell apoptosis was higher,the activity of Caspase-3 was higher,the expressions of shear Caspase-3 and Bax were higher and Bcl-2 was lower after HUVECs were treated with Hcy(P<0.05).The cell viability and migration were higher,cell apoptosis was lower,the activity of Caspase-3 was lower,the expressions of shear Caspase-3 and Bax were lower and Bcl-2 was higher after HUVECs were pretreated with different concentrations of selenium(P,0.05).Conclusion Selenium can inhibit homocysteine-induced endothelial cell dysfunction.
