摘 要:目的研究阿托伐他汀对糖尿病心肌病大鼠心肌组织LOX、NF-κB及MMP-2水平的影响。方法选取8周龄雌性健康大鼠25只,以随机数表法分为模型组、A组、B组、C组、空白组,每组5只。模型组与空白组给予生理盐水10 mg/(kg·d)灌胃,A、B、C组分别给予阿托伐他汀10、20、40 mg/(kg·d)灌胃。观察各组LVEDd、LVESd、LVEF、LOX、NF-κB、MMP-2、P38、Glu、TG及TC。结果模型组、A组、B组、C组、空白组LVEDd、LVESd呈降低趋势,且模型组>A组>B组>C组(P<0.05);模型组、A组、B组、C组、空白组LVEF呈上升趋势,且模型组A组>B组>C组,差异具有统计学意义(P<0.05)。五组大鼠TG、TC水平比较,模型组>A组>B组>C组(P<0.05);五组Glu水平比较,差异无统计学意义(P>0.05)。结论阿托伐他汀可降低LOX、NF-κB、MMP-2、P38及血脂水平,且随着剂量上升有增强趋势,能有效改善心功能,抑制心肌纤维化,但其降低空腹血糖的作用不明显。Objective To study the effects of atorvastatin on the levels of LOX, NF-κB and MMP-2 in myocardium of diabetic cardiomyopathy rats. Methods Twenty-five healthy female rats aged 8 weeks were selected and divided into model group, group A, group B, group C and blank group by random number table method, with 5 rats in each group.The model group and blank group were given 10 mg/(kg·d) of normal saline, and the groups A, B, C were given 10, 20,40 mg/(kg·d) of atorvastatin. The LVEDd, LVESd, LVEF, LOX, NF-κB, MMP-2, P38, Glu, TG and TC in each group were observed. Results The LVEDd and LVESd in the model group, group A, group B, group C and blank group decreased,and the model group>group A>group B>group C(P<0.05);the LVEF in the model group, group A, group B, group C and blank group increased, and the model groupgroup A>group B>group C( P<0.05). Com pared the TG and TC levels of the five groups, the model group >group A >group B >group C( P <0.05);there was no significant difference in the level of Glu among the five groups(P>0.05). Conclusion Atorvastatin can reduce the levels of LOX,NF-κB, MMP-2, P38 and blood lipid, and it is more significant with the increase of dose, atorvastatin can improve cardiac function effectively and inhibit myocardial fibrosis effectively, but it has no obvious effect on reducing fasting blood sugar.
