摘  要:目的:通过电针刺激印堂、百会穴对慢性不可预见性温和应激(CUMS)抑郁模型大鼠海马、中缝核miRNA-16及5-羟色胺转运体(SERT)的影响研究,揭示电针治疗抑郁症的可能作用机制。方法:雄性SD大鼠40只,随机分为4组,每组10只,分别为正常对照组、模型组、氟西汀组、电针组。除正常对照组外,其余各组大鼠进行孤养,并接受CUMS刺激方式造模。各组大鼠分别于应激前1天、应激后1天进行强迫游泳试验并记录数据,观察大鼠的行为学变化。运用RT-RCR技术检测各组大鼠海马、中缝核miRNA-16的表达变化;Western blot法观测各组大鼠海马、中缝核SERT蛋白的表达水平。结果:与正常对照组比较,模型组强迫游泳时被动漂浮时间延长(P<0.01),大鼠提前表现绝望;与模型组比较,氟西汀组和电针组被动漂浮时间缩短(P<0.05),表现较活跃;与氟西汀组比较,电针组漂浮时间较短(P<0.05)。与正常对照组比较,模型组大鼠海马、中缝核miRNA-16含量升高(P<0.01),SERT表达水平亦升高(P<0.01);与模型组比较,电针组海马和中缝核miRNA-16含量降低(P<0.05),SERT含量亦降低(P<0.05),差异均有统计学意义。结论:脑区miRNA-16的变化可能与抑郁症的发生相关,而电针能通过抑制脑区SERT的表达,发挥抗抑郁作用。Objective:To study the effects of electro-needling Yintang(EX-HN3)and Baihui(GV20)on miRNA-16 and SERT in hippocampus and raphe nucleus of rats with depression due to chronic unpredictable mild stress(CUMS),thus to reveal the possible mechanism of electro-acupuncture in the treatment of depression.Methods:40 SD rats were randomly divided into the normal cotrol group,the model group,the Fluoxetine group and the electro-acupuncture group,with 10 rats in each group.The depression model was established by CUMS and separation.One day before and after stress stimulation,the rats in each group were subjected to forced swimming test,and the behavioral changes of the rats were observed.The expressions of miRNA-16 and SERT protein in hippocampus and raphe nucleus of were respectively detected by RT-RCR technique and western blot method.Results:Compared to the normal control group,the passive floating time of the forced swimming test was prolonged in the model group(P<0.01),and the rats showed despair earlier.Compared to the model group,the passive floating time was shortened in the Fluoxetine group and in the electro-acupuncture group(P<0.05),and the rats were active.Compared to the Fluoxetine group,the passive floating time was shortened in the electro-acupuncture group(P<0.05).Compared to the normal control group,the miRNA-16 contents in the hippocampus and raphe nucleus were increased(P<0.01)and the SERT protein were also increased in the model group(P<0.01).Compared to the model group,the miRNA-16 contents were decreased(P<0.05)and the SERT protein were also decreased in the electro-acupuncture group(P<0.05).Conclusion:The expression levels of miRNA-16 in the brain may associate with the development of depression,and electro-acupuncture can exert anti-depressant effects by inhibiting the expression of SERT in the brain.
