摘 要:目的探讨血必净注射液对小鼠血液和肺组织中左氧氟沙星(LVFX)分布的影响。方法将108只小鼠随机分为实验组(血必净注射液联合LVFX)和对照组(0.9%氯化钠注射液),各54只。两组小鼠分别腹腔注射血必净注射液(22 mL/kg)或等体积0.9%氯化钠注射液,末次腹腔注射1 h后两组小鼠均静脉注射LVFX(132 mg/kg,1.32%);静脉注射3,5,10,30,60,120,240,480,720 min后,采集小鼠血液和肺组织,制成血浆或肺组织匀浆,以高效液相色谱法检测样本中LVFX质量浓度,采用DAS 2.0药代动力学分析软件计算药代动力学参数。结果对照组和实验组小鼠静脉注射LVFX后血液和肺组织中药时曲线均符合二室模型。对照组和实验组小鼠血液AUC0→12h分别为(223.57±23.42)mg/(L·h)和(192.02±20.34)mg/(L·h),小鼠肺组织中AUC0→12h分别为(218.48±31.05)mg/(L·h)和(283.66±21.53)mg/(L·h),AUC0→∞分别为(246.32±34.95)mg/(h·L)和(305.78±25.66)mg/(L·h),P均小于0.05)。结论血必净注射液可降低LVFX在血液中的质量浓度,升高其在肺组织中的质量浓度。Objective To investigate the effect of Xuebijing Injection on the distribution of levofloxacin in plasma and lung tissue of mice.Methods Totally 108 mice were randomly divided into the experimental group(Xuebijing Injection combined with levofloxacin)and the control group(0.9% Sodium Chloride Injection),54 mice in each group.The mice in the experimental group were intraperitoneally injected with Xuebijing Injection(22 mL/kg),while the mice in the control group were intraperitoneally injected with equal volume of 0.9% Sodium Chloride Injection.One hour after the last intraperitoneal injection,the mice in the two groups were injected levofloxacin(132 mg/kg,1.32%)intravenously.After 3,5,10,30,60,120,240,480 and 720 min of intravenous injection,the blood and lung tissues of mice were collected to make plasma or lung tissue homogenate.The concentrations of levofloxacin in the samples were detected by HPLC,and the pharmacokinetic parameters were calculated by DAS 2.0 software.Results After intravenous injection of levofloxacin,the drug concentration-time curve of blood and lung tissue of mice in the control group and the experimental group were consistent with the two-compartment model.The AUC0→12 h of the blood in the control group and the experimental group were(223.57±23.42)mg/(L·h)and(192.02±20.34)mg/(L·h),the AUC0→12 hof the lung tissue in the control group and the experimental group were(218.48±31.05)mg/(L·h)and(283.66±21.53)mg/(L·h),and the AUC0→∞of the lung tissue in the control group and the experimental group were(246.32±34.95)mg/(L·h)and(305.78±25.66)mg/(L·h)(P<0.05).Conclusion Xuebijing Injection can reduce the concentration of levofloxacin in blood and increase the concentration of levofloxacin in lung tissue.
