摘　　要：目的:研究河蟹肝胰腺中C-8神经酰胺-1-磷酸、N-十二烷醇-L-高丝氨酸内酯、磷酰胆碱和棕榈酰肉碱对细胞生长和亚细胞结构的影响.方法:利用细胞计数试剂盒和乳酸脱氢酶试验,检测不同质量浓度的4种脂类对斑马鱼胚胎成纤维细胞所表现的细胞增殖和细胞代谢的影响,并通过溶酶体和线粒体染色分析4种脂类对细胞溶酶体和线粒体形态结构的影响.结果:N-十二烷醇-L-高丝氨酸内酯表现为促进细胞增殖,并保持线粒体和溶酶体形态结构的完整;C-8神经酰胺-1-磷酸、磷酰胆碱、棕榈酰肉碱表现为抑制细胞增殖,并引起线粒体和溶酶体的结构发生不同程度的改变.结论:河蟹肝胰腺中的4种脂类物质不仅是食物的营养成分,还具有明显生物活性,能影响细胞代谢和亚细胞结构的完整性.Objective:To investigate the effects of C-8 Ceramide-1-phosphate,N-dodecanol-L-Homoserine lactone,Oleyloxyethyl Phosphorylcholine and Palmitoyl-L-carnitine from hepatopancreas of crabs on cell growth and subcellular structures.Methods:The effect of the four lipids on cell growth and metabolism of zebrafish embryonic fibroblasts at different concentrations were measured by using cell counting Kit and lactate dehydrogenase assay.The influence of the four lipids on morphology and structure of lysosomes and mitochondria were examined by fluorescence staining in PAC2 cells.Results:N-dodecanol-L-Homoserine lactone promoted cell proliferation and maintained the morphological and structural integrity of mitochondria and lysosomes.Conversely,C-8 Ceramide-1-phosphate,Oleyloxyethyl Phosphorylcholine and Palmitoyl-L-carnitine inhibited cell proliferation and altered the structure of mitochondria and lysosomes to different extent.Conclusion:The results indicated that the four lipids from hepatopancreas of crabs are not only food nutrients but also of obvious bioactivities on cell metabolism and subcellular structural integrity.
