摘　　要：目的:建立蒙药紫花高乌头药材HPLC指纹图谱及生物碱含量测定方法,用于不同产地紫花高乌头药材的质量评价。方法:采用Phenomenex Gemini C18色谱柱(250 mm×4.6 mm,5μm),以乙腈-0.04mol·L-1醋酸铵溶液为流动相进行梯度洗脱建立紫花高乌头药材的指纹图谱,并测定高乌甲素与冉乌头碱含量;采用相似度评价、聚类分析(CA)和主成分分析(PCA)3种方法对10批紫花高乌头药材指纹图谱进行研究。结果:建立了紫花高乌头药材HPLC指纹图谱,确定了14个共有峰,测定了10批次样品中高乌甲素与冉乌头碱含量;通过相似度评价、聚类分析及主成分分析结果可知10批紫花高乌头药材质量有差异,栽培品和野生品种之间质量有明显差异。结论:建立的HPLC指纹图谱结合含量测定,PCA和CA方法可以客观、有效、全面地用于紫花高乌头药材的质量评价。Objective:To establish a method of fingerprint analysis and determination of alkaloids in Aconitum excelsum Reichb by high performance liquid chromatography(HPLC)and to evaluate the quality of Aconitum excelsum Reichb in different regions.Methods:HPLC analysis was performed on an Phenomenex Gemini C18(250 mm×4.6 mm,5μm)with the mobile phase consisting of acetonitrile-0.04 mol·L-1 ammonium acetate solution for gradient elution to establish fingerprint for Aconitum excelsum Reichb and determine the contents of lappaconitine and ranaconitine.Similarity analysis,hierarchical clustering analysis(CA)and principal component analysis(PCA)were conducted to investigate fingerprints of 10 batches of Aconitum excelsum Reichb.Results:HPLC fingerprint of Aconitum excelsum Reichb was established and 14 common peaks were designated,and the contents of lappaconitine and ranaconitine were determined.The results showed that the quality of 10 batches of samples was not stable.There are significant differences between samples collected from the wild and those cultivated.Conclusion:The HPLC fingerprint,PCA,CA method can evaluate the quality of Aconitum excelsum Reichb objectively,effectively and thoroughly.
