摘 要:目的研究三七总皂苷(PNS)在缺糖缺氧-再灌注(OGD-R)下对新生大鼠海马神经干细胞(NSCs)的保护作用,探讨其可能机制。方法将生长状态良好的海马NSCs分为3组:对照组、模型组和实验组。对照组加有糖Earle氏液培养;模型组加无糖Earle氏液经OGD-R处理;实验组加无糖Earle氏液,在OGD-R基础上加PNS(15μg·mL-1)进行干预。以噻唑蓝法测定细胞存活率;以4’,6-二脒基-2-苯基吲哚(DAPI)检测细胞凋亡(阳性细胞数)情况;用免疫印迹法检测NSCs中卷曲蛋白1、散乱蛋白1和周期蛋白D1的表达量。结果对照组、模型组、实验组细胞的存活率分别为(100.00±0.00)%,(59.41±3.27)%和(83.35±11.52)%;上述这3组的阳性细胞数分别为248.41±26.61,117.54±14.86和192.25±21.13;上述这3组的卷曲蛋白1的相对表达水平分别为0.44±0.04,0.19±0.03和0.27±0.04,上述这3组的散乱蛋白1相对表达水平分别为0.43±0.04,0.20±0.04和0.30±0.03,上述这3组的周期蛋白D1蛋白相对表达水平分别为0.42±0.05,0.20±0.05和0.29±0.04。上述指标:模型组与对照组相比或者实验组与模型组相比,差异均有统计学意义(均P<0.05)。结论PNS能增加OGD-R新生大鼠海马NSCs的存活率,抑制NSCs的凋亡,其机制可能与激活Wnt/β-catenin信号通路有关。Objective To study the protective effect of Panax notoginseng saponins(PNS)on hippocampal neural stem cells(NSCs)in neonatal rats under hypoglycemia-hypoxia-reperfusion(OGD-R)and explore its possible mechanism.Methods The NSCs of the hippocampus with good growth were divided into three groups:control group(sugar Earle solution,normal culture),model group(sugar-free Earle solution,OGD-R),and experimental group(sugar-free Earle solution,PNS(15μg·mL-1)intervention culture based on OGD-R).The cell survival rate was detected by MTT.The cell apoptosis(positive cells)was measured by 4’,6-diamidyl-2-phenylindol(DAPI)staining.The expressions of curly protein 1,scattered protein 1 and cyclin D1 in NSCs were detected by Western blot.Results The cell survival rates in control group,model group and experimental group were(100.00±0.00)%,(59.41±3.27)%,(83.35±11.52)%,respectively;the positive cells in the above three groups were 248.41±26.61,117.54±14.86,192.25±21.13,respectively;the relative expression levels of curly protein 1 in the above three groups were0.44±0.04,0.19±0.03,0.27±0.04,respectively;the relative expression levels of scattered protein 1 in the above three groups were 0.43±0.04,0.20±0.04,0.30±0.03,respectively;the relative expression levels of cyclin D1 in the above three groups were 0.42±0.05,0.20±0.05,0.29±0.04,respectively.Comparing between model group and control group,or experimental group and model group,the differences of the factors were significant(all P<0.05).Conclusion PNS can promote the survival rates and inhibit apoptosis of NSCs in the hippocampus of OGD-R neonatal rats.The mechanism may be related to the activation of Wnt/β-catenin signaling pathway.
