摘  要:为了明晰营养因素是否同时影响内源性反转录病毒(Endogenous retrovirus, ERV)和免疫相关基因的表达,试验以采集的浙东白鹅肝脏样品为材料,采用荧光定量PCR方法测定了不同营养状态(禁食或脂肪酸处理)对鹅肝脏或肝细胞中两个ERV Pol序列(ERVK18P、ERVK25P)和免疫相关基因STAT3表达的影响。结果表明:与未禁食的70日龄浙东白鹅相比,禁食24 h会抑制肝脏中ERVK18P、ERVK25P和STAT3基因的表达;用亚油酸处理鹅原代肝细胞14 h,也会显著抑制ERVK18P、ERVK25P和STAT3基因的表达。说明营养因素可以影响ERV和免疫相关基因STAT3的表达,ERV可能是营养因素调控鹅肝脏中STAT3基因表达的潜在介导物。In order to clarify whether nutritional factors regulate the expression of Endogenous retrovirus(ERV)and immune-related genes.The liver samples of Zhedong white geese were collected as materials.The expression of two ERV Pol sequences(ERVK18P,ERVK25P)and an immune-related gene(STAT3)were determined in goose livers or hepatocytes under different nutritional conditions(starvation or fatty acid treatment)by quantitative PCR.The results showed that the expressions of ERVK18P,ERVK25P and STAT3 genes were all inhibited in the livers of Zhedong white geese starved for 24 h compared with those 70-day-old Zhedong white geese not starved.The expressions of ERVK18P,ERVK25P and STAT3 genes were also significantly inhibited by linoleic acid treatment for 14 h.It indicated that nutritional factors could regulate the expressions of ERV and immune-related gene STAT3.ERV may be a potential mediator in regulation of nutritional factors on STAT3 expression in goose liver.
