摘  要:转铁蛋白(TF)是运输铁离子的重要蛋白,其与低氧有密切联系。研究比较了藏鸡在常氧(02,21%)和低氧(02,13%)孵化环境中胚胎尿囊绒毛膜(CAM)组织7T基因mRNA表达变化及其与茶花鸡的差异,以及在不同氧气浓度培养条件下鸡胚成纤维细胞(DF-1)中7T基因的mRNA表达变化,阐述7T基因表达与低氧适应的关系。结果显示:在鸡胚孵化期第11~16天,藏鸡CAM组织中7T基因在低氧环境下的表达高于常氧环境及茶花鸡(P<0.05),且低氧明显刺激DF-1细胞中rF基因的表达(P<0.05)。研究表明在低氧环境中藏鸡CAM组织7T基因表现出明显的增加,促进了血液中Fe>的运输以及增加血氧结合能力从而使其胚胎能更好的适应低氧环境。Transferrin(TF)is an important protein of transporting iron,which is closely related to hypoxia.In this study,we compared the expression of TF gene in Tibetan chicken embryonic chorioallantoic membrane(CAM)tissues with Chahua chicken in normoxia(O2,21%)and hypoxia(O2,13%)incubation environments,and compared the expression of TF gene in chicken embryonic fibroblast cell(DF-1)cultured in different oxygen concentration.The relationship between expression of TF gene and hypoxic adaptation were expounded.The results showed that the expression of TF gene in CAM tissues of Tibetan chickens in hypoxic incubation on the list to 16th day was higher than that in normoxia incubation and Chahua chicken,and hypoxia significantly stimulated the expression of TF gene in DF-1 cells.The results indicated that the expression of TF gene in CAM tissue of Tibetan chickens increased significantly in hypoxic environment,which promoted the transport of Fe^3+ in blood and increased the ability of blood oxygen binding,thus enhancing the ability of Tibetan chickens to adapt to hypoxic environment.
