摘 要:目的:研究寿胎丸对复发性流产(RSA)大鼠子宫蜕膜组织中各型水通道蛋白(AQP)表达的影响,初步筛选与RSA相关的水通道蛋白亚型,阐述其可能的安胎机制。方法:选择健康SPF级SD大鼠,将雌鼠与雄鼠2∶1合笼,按妊娠顺序随机分为寿胎丸高、中、低剂量组,地屈孕酮组,模型组及正常妊娠组。除正常妊娠组,其余各组孕鼠在妊娠第1~9天每天下午予羟基脲片水溶液灌胃(450 mg/kg),妊娠第10天早上予米非司酮片水溶液灌胃(4.0 mg/kg)。另外,妊娠第1~9天每日上午,寿胎丸高、中、低剂量组分别予寿胎丸中药液灌胃(2.0、1.0、0.5 g/kg);地屈孕酮组予地屈孕酮水溶液灌胃(3.02 mg/kg);模型组和正常妊娠组予等量0.9%氯化钠注射液灌胃。各组孕鼠均于末次给药24 h后断颈处死,无菌条件下解剖孕鼠取子宫蜕膜组织,采用蛋白印迹法检测各组孕鼠子宫蜕膜组织AQP的表达情况。结果:寿胎丸高、中、低剂量组及地屈孕酮组孕鼠流产率均较模型组降低,差异有统计学意义(P<0.05);蛋白印迹结果显示,与正常妊娠组相比,模型组孕鼠子宫蜕膜组织中AQP0、AQP12呈高表达,AQP1、AQP2、AQP3、AQP4、AQP5、AQP6、AQP8、AQP9、AQP10、AQP11呈低表达,差异有统计学意义(P<0.05),AQP7呈低表达,差异无统计学意义;进行药物干预后,与模型组相比,寿胎丸高、中、低剂量组及地屈孕酮组孕鼠蜕膜组织中AQP0表达水平降低,AQP1、AQP2、AQP3、AQP4、AQP5、AQP6、AQP8、AQP9、AQP10、AQP11表达水平升高,差异有统计学意义(P<0.05),AQP12表达无统计学意义。结论:寿胎丸能促进胚胎发育,有效降低RSA孕鼠的胚胎吸收率,可能通过下调蜕膜组织中AQP0的表达,上调AQP1、AQP2、AQP3、AQP4、AQP5、AQP6、AQP8、AQP9、AQP10、AQP11的表达,调节母胎界面水液平衡维持妊娠。Objective:To study the effect of Shoutai Pills on aquaporin expression of recurrent abortion model rat uterine decidua tissues and preliminarily screen aquaporin protein subtpyes associated with recurrent abortion and explain its possible mechanism.Method:Healthy SPF grade SD rats were selected. Sccording to the proportion of 2:1(female:male),the rats were put into the cage. The rats were randomly divided into high, medium and low doses groups of Shoutai Pills,dydrogesterone group,model group and normal pregnancy group according to the pregnency order.In addition to the normal gestation group, pregnant rats in the other groups were given hydroxyl urea tablets water solution(450 mg/kg) every afternoon on the 1st to 9thday of gestation, and mifepristone tablets water solution(4.0 mg/kg) on the morning of the 10th day of gestation.In addition,in the morning of the 1st to 9th the day of gestation,the high, medium and low doses groups of Shoutai Pills were given the drug solution by intragastric administration(2.0,1.0, 0.5 g/kg).The dydrogesterone group was given an aqueous solution to the stomach(3.02 mg/kg). The model group and the normal pregnancy group were given equal 0.9% sodium chloride injection by gavage.All the pregnant mice in each group were sacrificed after 24 h after the last administration, and their uterine decidua tissues were dissected under aseptic conditionsand the expression of aquaporins in uterine decidua tissues of pregnant mice in each group was detected by western bloting.Results:The abortion rate of pregnant rats in the high, medium and low doses groups of Shoutai Pills was lower than that in the model groupand the difference was statistically significant(P<0.05).Western bloting results showed that AQP0 and AQP12 were highly expressed in the uterine decidua tissue of pregnant rats in the model group compared with that of the normal pregnancy group, while AQP1, AQP2, AQP3, AQP4, AQP5, AQP6, AQP8, AQP9, AQP10 and AQP11 were low expressedwith statistically significant differences(P<0.05).After
