摘  要:目的探讨PCR-毛细管电泳法在检测小龙虾中沙门氏菌、副溶血性弧菌和金黄色葡萄球菌的应用效果。方法同时应用食品微生物检验国标GB 4789方法和PCR-毛细管电泳法检测60份小龙虾中的三种致病菌,对两种检测方法的效果进行比较评价。结果利用国标方法,小龙虾中沙门氏菌、金黄色葡萄球菌、副溶血性弧菌的检出率分别为25.00%、20.00%、10.00%。运用PCR-毛细管电泳方法,沙门氏菌、金黄色葡萄球菌、副溶血性弧菌的检出率分别为30.00%、20.00%、10.00%。PCR-毛细管电泳方法沙门氏菌的检出率高于国标方法,三种致病菌的检出情况与国标方法一致,检验过程可控制在24 h之内。结论 PCR-毛细管电泳方法可用于小龙虾中沙门氏菌、金黄色葡萄球菌、副溶血性弧菌的检测,具有快速、灵敏、自动化程度高等优点。Objective To elucidate the application prospect of PCR-capillary electrophoresis in the detection of salmonella,Vibrio parahaemolyticus and Staphylococcus aureus in crayfish.Methods Three types of pathogenic bacteria of 60 crayfish products were detected by GB4789 national food microbiological testing methods and the PCR-capillary electrophoresis method.Results The positive rates of salmonella,S.aureus and V.parahaemolyticus by the GB4789 national food microbiological testing methods were 25.00%,20.00%,and 10.00% respectively,and the detection rates of salmonella,S.aureus and V.parahaemolyticus by PCR-capillary electrophoresis were 30.00%,20.00%,and 10.00% respectively.The positive rate of salmonella by PCR-capillary electrophoresis was higher than that by the GB4789 national food microbiological testing methods,and the positive rates of S.aureus and V.parahaemolyticus by the two methods was coincident,and the inspection process could be controlled within 24 hours.Conclusion The PCR-capillary electrophoresis method can be used for detection of salmonella,S.aureus,and V.parahaemolyticus in crayfish,and it has the rapid and sensitive advantages,and higher degree of automation.
