摘 要:目的基于数据挖掘方法分析高迁移率族蛋白1(HMGB1)在健康人群及急性淋巴细胞白血病(ALL)患者体内的表达及意义。方法通过Oncomine v4.5数据库提取HMGB1在白血病和健康人群中转录水平的相关数据,对数据库中的基因表达信息进行荟萃分析,并通过KEGG PATHWAY分析HMGB1参与的重要信号通路。结果在443项关于HMGB1在各种恶性肿瘤患者与健康人群之间的对比研究中,有38项研究提示HMGB1在恶性肿瘤患者和健康人群中的表达水平差异有统计学意义(P<0.05),其中在白血病患者中表达增高的有3项。在271项关于HMGB1在多种不同恶性肿瘤患者之间表达情况的对比研究中,有15项研究提示HMGB1在不同恶性肿瘤患者中的表达差异有统计学意义(P<0.05),其中有4项提示HMGB1在白血病患者中的表达高于其他肿瘤患者。5项关于HMGB1在ALL与健康人群中表达情况的研究提示,HMGB1在ALL中的表达量显著高于健康人。KEGG PATHWAY通路分析提示,HMGB1主要参与了自噬、碱基切除修复、程序性坏死等通路。结论HMGB1在ALL中呈现高表达,且参与ALL的发生、发展,通过诱导自噬、DNA损伤修复等促进化疗药物耐药。Objective To investigate the expression and significance of high mobility group protein box 1(HMGB1)in acute lymphoblastic leukemia(ALL)and normal tissues based on data mining.Methods Expression data of HMGB1 in ALL and normal tissues were extracted from Oncomine v4.5 database.Crucial signaling pathways of HMGB1 in ALL were analyzed by KEGG PATHWAY analysis.Results A total of 38 of 443 studies about expression of HMGB1 in different cancers and healthy volunteers enrolled from Oncomine database were statistically different(P<0.05),with 3 highly expressed HMGB1 in leukemia.A total of 15 of 271 studies on H MGB1 expression among various of cancers were statistical different(P<0.05),and 4 of these studies were associated with leukemia.Totally 5 studies about HMGB1 expression levels in ALL patients and healthy volunteers were compared,HMGB1 expression in ALL patients were higher than healthy volunteers.KEGG PATHWAY analysis showed that HMGB1 was mainly involved in autophagy,base excision repair,necroptosis,etc.Conclusion HMGB1 is highly expressed in ALL,and its expression level is associated with the pathogenesis and chemotherapy resistance promotion of ALL by inducing autophagy and base excision repair.
