初沉污泥中真菌群落结构与病原真菌组成研究

摘  要:为调查初沉污泥的真菌多样性及其生物安全性,以郑州市某污水处理厂(活性污泥法)的初沉池污泥为研究对象,通过Illumina MiSeq高通量测序技术,分析真菌群落组成和病原真菌的含量.chao1指数显示,该污水处理厂1 a内初沉池污泥均显示出丰富的真菌多样性,并且冬季的真菌多样性略低于夏季的.PCA结果显示,1 a内初沉池污泥的真菌群落没有明显的季节变化趋势.Ascomycota,Basidiomycota在四季初沉污泥中均是优势真菌门.除了Ascomycota,Basidiomycota外,Zygomycota,Glomeromycota也在不同季节的初沉污泥中检测到.在门水平上的未分类真菌在四季初沉污泥中的丰度都很高.属水平分析结果显示,Trichosporon,Candida在四季初沉污泥中均是优势真菌属.在初沉池污泥中发现Trichosporon.Loubieri,Candida.spp等病原真菌.为避免造成环境二次污染,初沉池污泥在排出前需进行处理.With Illumina MiSeq high throughput sequencing technology,the article analyzes composition of fungal community and content of pathogenic Fungi by taking primary sludge in an activated sludge method municipal wastewater treat plant in Zhengzhou as researching object to investigate fungal diversity and biological safety of primary sludge.The chao1 index shows that,the fungal community structure of sludge in primary settling tank of this municipal sewage treatment plant showed abundant fungal diversity during a one-year cycle,and the fungal diversity of primary sludge in the winter is slightly lower than that in the summer.PCA result shows that,there was no obvious seasonal variation trend for fungal community in the sludge of primary settling pond during one year cycle.Both Ascomycota and Basidiomycota are the dominant fungus phylum in four seasonal primary settling sludge.Other than Ascomycota and Basidiomycota,Zygomycota and Glomeromycota are also detected in primary settling sludge in different seasons.It is noteworthy that unclassified fungi at phylum level is high in abundance in four seasonal primary sludge.The analysis results at the genus level show that,Trichosporon and Candida are dominant fungi in four seasonal primary sludge.In addition,the Pathogenic Fungi of Trichosporon,Loubieri and Candida.spp are found in primary sludge.In order to avoid secondary environmental pollution,primary sludge needs further treatment before it is discharged into the environment.
