摘 要:目的观察霍山石斛对荷宫颈癌SiHa细胞小鼠免疫功能的影响。方法将60只BALB/c裸鼠随机均分为石斛高、低剂量组和模型组,于腋窝皮下注射宫颈癌SiHa细胞造模。石斛高、低剂量组大鼠ig给予相应剂量的药物,模型组ig给予等量生理盐水。称量肿瘤组织大小及重量;采用流式细胞术检测荷瘤小鼠外周血CD8^+T细胞计数;免疫荧光染色检测肿瘤组织中CD8^+T细胞的表达情况;Western Blot法检测肿瘤组织中IFN-γ的表达水平。结果石斛高、低剂量组肿瘤组织的重量明显低于模型组的;石斛高剂量组肿瘤组织的重量明显低于低剂量组的,差异具统计学意义。石斛高、低剂量组小鼠外周血CD8^+T细胞占CD3^+T的比例均高于模型组的。石斛高、低剂量组荷瘤小鼠肿瘤组织中CD8+T细胞的表达及IFN-γ的分泌水平均高于模型组的。结论霍山石斛能抑制荷SiHa宫颈癌小鼠肿瘤组织的生长,调节外周血CD8+T细胞的数量及影响细胞因子IFN-γ的分泌。OBJECTIVE To observe the effect of Dendrobium huoshanense(DH)on the immune function in SiHa tumor-bearing mice.METHODS 60 BALB/c nude mices modelled by subcutaneous injection of SiHa cells were divided into 3 groups randomly:model group,high dose of DH group and low dose of DH group.Mice in high and low dose of DH groups were intragastrically administrated with DH,while the mice in model group were given normal saline.Then weight of the tumor tissue was detected.The CD8^+T cell counts in peripheral blood of SiHa mice were compared by Flow cytometry.The expression of CD8^+T cells in tumor tissue was tested by Immunofluorescence staining.The expression of IFN-γin tumor tissue was examined by Western Blot.RESULTS Tumor volume(maximum diameter)and mass of high and low dose of DH group was lower than the model group significantly.Compared with the control group,the CD8^+T cell counts in peripheral blood of SiHa mice and the expressions of CD8^+T and IFN-γin tumor tissue of high and low dose of DH group were significantly higher.CONCLUSION DH can inhibit the growth of tumor tissues in mice with cervical cancer SiHa cells and regulate the number and expression of CD8^+T cells,affect the secretion of IFN-γ.
