摘 要:目的:研究加味丹参饮通过Notch信号通路对心肌缺血损伤模型大鼠的保护作用。方法:20只大鼠建立心肌缺血损伤模型,分为假手术组、模型组、美托洛尔组和加味丹参饮,各5只。检测心功能心率(HR)、左心室收缩压(LVSP)、左心室舒张末压(LVEDP)、舒张压、收缩压、心肌梗死面积、心肌缺血百分率、活性氧水平,病理学观察,采用酶联免疫吸附试验检测丙二醛(Malondialdehyde,MDA)、超氧化物歧化酶(Superoxide dismutase,SOD)、谷胱甘肽过氧化物酶(Glutathione peroxidase,GSH-Px)、乳酸脱氢酶(lactate dehydrogenase,LDH)和肌酸激酶(Creatine kinase,CK)水平,Western blot检测Notch信号通路相关蛋白Notch1、Hes1、缺氧诱导因子1α(Hypoxia inducible factor 1alpha,HIF-1α)表达。结果:与模型组相比,加味丹参饮组HR、LVSP、收缩压、舒张压、SOD、GSH-Px升高,LVEDP、心肌梗死面积、心肌缺血百分率、活性氧水平、MDA、LDH、CK降低(P<0.05),Notch1、Hes1、HIF-1α表达降低(P<0.05)。结论:加味丹参饮通过抑制Notch信号通路相关蛋白表达,改善大鼠氧化应激反应,从而对心肌缺血损伤大鼠起到一定的保护作用。Objective:To study the protective effect of Jiawei Danshen Yin on myocardial ischemia injury model rats through Notch signaling pathway. Methods:Twenty rats were divided into sham operation group,model group,metoprolol group and modified Salvia miltiorrhiza drink 5 rats each. Heart function heart rate(HR),left ventricular systolic pressure(LVSP),left ventricular end-diastolic pressure(LVEDP),diastolic pressure,systolic pressure,infarct size,myocardial ischemia percentage,reactive oxygen species level were measured. Pathological observation. Malondialdehyde(MDA),superoxide dismutase(SOD),glutathione peroxidase(Glu)were detected by enzyme-linked immunosorbent assay. Tathione peroxidase(GSH-Px),lactate dehydrogenase(LDH)and creatine kinase(CK)levels were detected. Western blot was used to detect the expression of Notch signaling pathway related proteins Notch1,Hes1 and hypoxia inducible factor 1 α(HIF-1α). Results:Compared with the model group,HR,LVSP,systolic blood pressure,diastolic blood pressure,SOD,GSH-Px increased,LVEDP,myocardial infarction area,myocardial ischemia percentage,reactive oxygen species level,MDA,LDH,CK decreased(P<0.05),Notch1,Hes1,HIF-1 a expression decreased(P<0.05). Conclusion:Jiawei Danshen Yin can improve oxidative stress response by inhibiting Notch signaling pathway-related protein expression,thus playing a protective role in rats with myocardial ischemia injury.
