摘 要:目的:建立经典方"桔梗汤"工艺优化质量评价方法。方法:通过正交试验设计优化工艺、高效液相色谱法进行甘草苷、甘草酸、桔梗皂苷D含量测定,并建立相关特征图谱以确定工艺优化效果。结果:在回流提取3次,每次加水倍量为20倍,每次提取时间为1 h时提取效果最好。含量测定表明工艺优化后桔梗汤中甘草酸、甘草苷、桔梗皂苷D含量较标准煎液均有所提高;该方法精密度高,稳定性较好,阴性无干扰;特征图谱方法考察结果,该方法稳定性RSD%为1.24%、重复性RSD%为1.92%,可用来作为桔梗汤质量控制标准之一。结论:该实验对桔梗汤提取工艺及色谱特征进行了相关研究,优化桔梗汤提取工艺,为桔梗汤复方的工业化提供理论基础;为进一步的研究提供科学依据。Objective:To establish a method for improving the process and quality evaluation of theclassic Campanulaceae decoction. Methods:The content of glycyrrhizin,glycyrrhizin and Platycodin D of Campanulaceae were determined by TLC and HPLC,and related characteristics were established. Process improvement is carried out through orthogonal design. Results:The samples on the thin plate and the cross-references had the same color spots in the corresponding positions of Campanulaceae saponin D and glycyrrhizic acid. The content of glycyrrhizin,glycyrrhizin and Platycodin D in the chromatogram are in accordance with the National Pharmacopoeia standards;The method has high precision,good stability and no interference. The results of the feature map method show that the stability RSD% is 1.24% and the repeatability RSD% is 1.92%. It can be used as one of the quality control standards of Campanulaceae decoction. Conclusion:This experiment studied the characteristics of Campanulaceae decoction,optimized the extraction technology of Campanulaceae decoction,and provided the theoretical basis for the industrialization of Campanulaceae decoction,provide scientific basis for further research.
