摘  要:目的:探讨白背桐黄钻方调控失眠模型大鼠脑电活动、神经递质、炎症因子和氧化应激的促眠机制。方法:小平台水环境法剥夺大鼠睡眠复制失眠动物模型,以右佐匹克隆为对照,将80只大鼠随机分为空白组、模型组、右佐匹克组和瑶药组,造模结束后,记录各组大鼠的全身变化,采用脑电图仪检测脑电活动[觉醒(Wake)、慢波睡眠I期(SWS1)、慢波睡眠Ⅱ期(SWS2)和快动眼睡眠(REMS)四个时相],分别采用酶联免疫法(ELISA)检测外周血和Western blot检测脑组织的神经递质[5-羟色胺(5-hydroxytryptamine,5-HT)]及炎症因子[白细胞介素-1(interleukin-1,IL-1)、白细胞介素-6(interleukin-6,IL-6)、肿瘤坏死因子(tumor necrosis factor alpha,TNF-α],采用分光光度计检测脑干的一氧化氮(nitric oxide,NO)、超氧化物歧化酶(superoxide dismutase,SOD)和丙二醛(malondialdehyde,MDA)。结果:空白组大鼠精神状态良好,活动机灵,反应敏捷,毛发有光泽,体重明显增加。模型组大鼠各项均减弱,瑶药组及右佐匹克组大鼠精神状态、活动及反应情况不如空白组大鼠,体重增长较空白组慢。与空白组比,模型组大鼠体重增加量、总睡眠时间、外周血和脑组织的5-HT、IL-1、IL-6和TNF-α水平、NREMS比例、脑干SOD水平均明显降低,睡眠潜伏期延长、觉醒比例增加,脑干NO和MDA增加(P<0.05)。与模型组比,瑶药组及右佐匹克组大鼠体重增加量、总睡眠时间、外周血和脑组织5-HT、IL-1、IL-6和TNF-α及脑干SOD水平升高明显,睡眠潜伏期缩短,觉醒比例降低明显,脑干NO和MDA降低(P<0.05);瑶药组及右佐匹克组比较均无统计学意义(P>0.05)。结论:瑶药白背桐黄钻方可较好的改善失眠模型大鼠的全身症状和睡眠状态,有效控制大鼠体重减轻,其机制可能与其可调控外周血和脑组织的神经递质、炎症因子及氧化应激有关。Objective:To explore the hypnogenetic mechanism underlying regulation of electrical activity,neurotransmitter,inflammatory factors and oxidative stress in insomnia model rats by Baibeitong Huangzhuan Formula.Methods:We established insomnia rat models using the small platform with water environment method.We divided 80 rats randomL y into 4 groups:the blank group,model group,dexzopiclone group and Baibeitong Huangzhuan Formula group.The changes in the whole body and the electrical activity of the brain(Wake,SWS1,SWS2 and REMS)were detected by electroencephalograph,neurotransmitter[5-hydroxytryptamine,5-HT)]and inflammatory factors[interleukin-1(L-1),interleukin-6(IL-6),tumor necrosis factor alpha(TNF-α)],nitric oxide(NO),superoxide dismutase(SOD)and malondialdehyde(MDA)in peripheral blood or brain tissue detected by ELISA or Western blot were observed.Results:Rats in the blank group were good in mental state,activity,responsiveness,lustrous-hair,and their body weight increased significantly.Rats in the model group were poor in all respects.As compared with rats in the blank group,rats in the Baibeitong Huangzhuan Formula group and dexzopiclone groups had poorer mental state,activity and responsiveness and their body weight increased more slowly.Rats in the model group were significantly decreased in terms of weight gain,total sleep time,the contents of 5-HT,IL-1,IL-6,TNF-αand SOD in peripheral blood or brain tissue,NREMS ratio,sleep latency extended,awakening ratio,NO and MDA had increased(P<0.05).Rats in the Baibeitong Huangzhuan Formula group and dexzopiclone group had markedly increased in terms of weight gain,total sleep time,the contents of 5-HT,IL-1,IL-6,TNF-αand SOD in peripheral blood or brain tissue and sleep latency shortened,awakening ratio,NO and MDA had reduced compared with those of the model group(P<0.05).No statistically differences were found between the Baibeitong Huangzhuan Formula group and dexzopiclone group.Conclusion:Baibeitong Huangzhuan Formula is helpful in improving general symptom an
