高效液相色谱指纹图谱法分析阿胶强骨口服液标志成分变化

摘　　要：建立了阿胶强骨口服液中毛蕊异黄酮葡萄糖苷和毛蕊花糖苷的高效液相色谱指纹图谱含量测定方法,并以其作为检测指标对指纹图谱进行相似度评价。采用Welch Ultimate XB C18色谱柱(4.6 mm×250 mm,5μm),以甲醇-0.2%磷酸溶液为流动相,流速1.0 mL·min^-1,进样量10μL,检测波长305 nm,柱温30℃,测定了阿胶强骨口服液的高效液相色谱指纹图谱,确认了9个共有峰,10批阿胶强骨口服液指纹图谱相似度均在0.920以上;4批阿胶强骨口服液中毛蕊异黄酮葡萄糖苷和毛蕊花糖苷的含量无显著性差异(P>0.05)。所建立的高效液相色谱指纹图谱含量测定方法,结果准确,稳定可行,为阿胶强骨口服液质量控制提供了有效途径。We established HPLC fingerprint method for the determination of calycosin 7-O-β-D-glucopyranoside and verbascoside in Ejiao Qianggu Oral Liquid,which are used as detection indexes for similarity evaluation of fingerprints.The HPLC analysis is performed on a Welch Ultimate XB C18 column(4.6 mm×250 mm,5μm),in which the mobile phase consists of methanol-0.2%phosphoric acid,the flow rate is 1.0 mL·min^-1,the injection volume is 10μL,the detection wavelength is 305 nm,and the column temperature is 30℃.The HPLC fingerprint of Ejiao Qianggu Oral Liquid is determined,and 9 common chromatographic peaks are identified.The similarity values of fingerprints of 10 batches of Ejiao Qianggu Oral Liquid are all above 0.920.There is no significant difference in the determination of the contents of calycosin 7-O-β-D-glucopyranoside and verbascoside in 4 batches of Ejiao Qianggu Oral Liquid(P>0.05).The established HPLC fingerprint method is accurate,stable,and feasible,which provides an effective way for the quality control of Ejiao Qianggu Oral Liquid.
