摘 要:目的探讨白介素10(IC-10)对人成熟树突状细胞(mDCs)F-ccUn结构及其结合蛋白表达情况的影响&方法从新鲜的人外周血分选出CD141单核细胞,诱导培养成mDCs,用10 ng/ml IC-10处理48 h,利用激光共聚焦显微镜观察mDCs细胞骨架结构的变化,并通过Western b/t实验和免疫荧光实验分析IC-10对mDCs细胞骨架结合蛋白Fas-cin-1 Cofilin和磷酸化Cofilin(p-Cofilin)的表达及其定位的影响。结果IL-IO处理mDCs后,其F-actin含量明显上升,细胞突起增加,Fascin-1和p-Cofilin的表达水平上调,而且3种细胞骨架结合蛋白的定位均发生改变。结论IL-10能够影响mDCs细胞骨及其合的定位,细胞骨架结构发生重组,为研究IL-IO对mDCs免疫学功能的影响奠定了基础.Objective To study the effects of interleukin-10(IL-10)on the F-actin strueture and its binding pro-teins of mature dendritic ells(mDCs).Methods CD14+monocytes were separated from fresh human peripheral blood,and then induced into mDCs in vitro.Subsequently,mDCs were treated with 10 ng/ml IL-10 for48 h.The changes of mDC cytoskeleton structure were observed using confocal laser scanning microscope.In addition,the effcts of IL-10 the expression and location of cytoskeletal binding proteins in mDCs,including Fascin-1,Cofilin,and phosphorylated Cofilin(p-Cofilin)were analyzed by Westerm blot and immunfluorescence assay.Results The results showed that the content of F-actin and the density of filopodia was dramatically increased.The expres-sion levels of Fascin-l and p-Cofilin were up-regulated.At the same time,the location of three cytoskeletal binding proteins was changed.Conclusion In conclusion,IL-10 can affeet F-actin structure of mDCs and the expression and location of cytoskeletal binding protein,which indicates that the cytoskeleton remodeling is occured in re-sponse to IL-10.It lays the foundation for studying the effects of IL-10 on the immune function of mDCs.
