摘 要:目的:研究观察消痰化瘀利窍方对慢性间歇性低氧(CIH)暴露大鼠肠系膜动脉纤维化的改善作用,并探讨其可能的分子机制。方法:大鼠随机分为3组:常氧对照组(Normoxia)、慢性间歇性低氧暴露组(CIH)、中药干预+慢性间歇性低氧暴露组(Formula+CIH)。Normoxia组暴露于常氧环境,CIH与Formula+CIH组暴露于间歇性低氧环境。其中,Formula+CIH组于每日造模前中药水煎液灌胃(24 g/kg),而CIH组与Normoxia组给予同等体积生理盐水。造模结束后,应用天狼星红染色观察各组大鼠肠系膜动脉纤维化水平,通过Western blot检测I型胶原(Col-I)蛋白和III型胶原(Col-III)蛋白的表达,应用q-PCR技术测定TGF-β和c-JUN的mRNA水平。结果:与Normoxia组相比,CIH组暴露大鼠肠系膜动脉出现显著的纤维化,Col-I、Col-III蛋白表达显著升高,伴随TGF-β和c-JUN的mRNA水平明显上调;消痰化瘀利窍方干预能够显著降低CIH暴露大鼠肠系膜动脉纤维化水平,抑制Col-I、Col-III以及TGF-β和c-JUN的表达水平。结论:消痰化瘀利窍方能够显著改善CIH暴露大鼠肠系膜动脉纤维化。Objective: To observe the effect of Xiaotan Huayu Liqiao Formula on mesenteric artery fibrosis in rats exposed to chronic intermittent hypoxia (CIH) and explore its possible molecular mechanism. Methods: The rats were randomly divided into three groups: normoxia control group, CIH group, and Formula + CIH group. Normoxia group was exposed to normoxic environment, while CIH and Formula + CIH group were exposed to intermittent hypoxia environment. In the Formula + CIH group, the decoction of Chinese medicine was infused into the stomach (24 g / kg) before modeling every day, while the same volume of normal saline was given to the CIH group and the normoxia group. Then, picrosirius staining was used to observe the level of mesenteric artery fibrosis, Wes-tern blot was used to detect the expression of type I collagen (CoL-I) protein and type III collagen (Col-III) protein, and the mRNA levels of TGF-β and c-JUN were measured by q-PCR. Results: compared with normoxia group, the mesenteric artery in CIH group showed significant fibrosis, the expression of CoL-I and Col-III protein was significantly increased, and the mRNA levels of TGF-β and c-JUN were significantly increased;the intervention of Xiaotan Huayu Liqiao Formula could significantly reduce the level of mesenteric artery fibrosis and the expression of CoL-I, Col-III, TGF-β and c- JUN. Conclusion: Xiaotan Huayu Liqiao Formula can significantly improve the fibrosis of mesenteric artery in rats exposed to CIH.
