摘  要:为了明确巴西蕉与香蕉枯萎病菌互作过程中WRKY转录因子的作用,对巴西蕉与香蕉枯萎病菌互作2 d的转录组中WRKY基因的表达量进行分析。结果表明,在互作过程中筛选到log值大于1的基因有55个,1个基因是上调表达的,其余是下调表达。55个基因中第Ⅰ类家族包含5个基因;Ⅱa亚家族中包含8个基因,Ⅱb亚家族中包含5个基因,Ⅱc亚家族中包含13个基因,Ⅱd亚家族中包含7个基因,Ⅱe亚家族中包含3个基因;第Ⅲ类家族中包含14个基因。55个基因在根中的表达量均大于在叶和果中的表达量。选取MaWRKY003、MaWRKY010、MaWRKY063和MaWRKY075四个差异显著的基因进行荧光定量PCR分析,结果表明与转录组数据结果趋势一致。对4个基因的启动子进行分析,发现4个基因启动子中所包含的元件不同,MaWRKY003、Ma WRKY063和MaWRKY075基因启动子中含有响应与病相关的激素响应元件,而MaWRKY010基因启动子中没有响应与病相关的响应元件,推测MaWRKY003、MaWRKY063和MaWRKY075三个基因可能是通过激素途径来响应香蕉枯萎病菌,而MaWRKY010可能是通过其他途径来响应香蕉枯萎病菌。以上结果表明MaWRKYs基因在巴西蕉与枯萎病菌互作过程中下调表达可能是巴西蕉易感枯萎病的原因之一。The expression of M a WRKYs were analyzed in the 2 d transcriptome in order to clarify the role of WRKY transcription factor between banana and Fusarium oxysporum f.sp cubense.The results showed that 55 MaWRKYs with log value greater than 1 were screened during the interaction,one gene was up-regulated and the others were down-regulated.Among these 55 MaWRKYs,classⅠhad 5 MaWRKYs,Ⅱa subfamily had 8 MaWRKYs,Ⅱb subfamily had 5 MaWRKYs,Ⅱc subfamily had 13 MaWRKYs,Ⅱd subfamily had 7 MaWRKYs andⅡe subfamily had 3 MaWRKYs,classⅢhad 14 MaWRKYs.The expression level of 55 MaWRKYs in root was higher than that in leaf and fruit.The expression of MaWRKY003,MaWRKY010,MaWRKY063 and MaWRKY075 were analyzed by fluorescence quantitative PCR and the results were consistent with the transcriptome data.The promoters of the four genes were analyzed,MaWRKY003,MaWRKY063 and MaWRKY075 promoters had hormone response elements relating to the disease,while MaWRKY010 promoter had no response elements relating to the disease.It is speculated that MaWRKY003,MaWRKY063 and MaWRKY075 may respond to Fusarium oxysporum f.sp cubense through hormone pathways,while MaWRKY010 may respond to Fusarium oxysporum f.sp cubense through other pathways.These results showed that the down-regulated expression of MaWRKYs in the interaction between banana and Fusarium oxysporum f.sp cubense might be one of the reasons for banana susceptibility to fusarium wilt of banana.
