摘  要:分离和分析花魔芋的抗病基因同源序列(resistance gene analogs,RGAs),为魔芋抗病相关基因的克隆和分析提供了机遇。本研究利用生物信息学手段,参考已知植物抗病基因的NBS-LRR类保守结构域筛选并合成了4对简并引物,从花魔芋的基因组DNA中扩增出4条NBS-LRR类抗病基因同源序列。序列分析表明,这4条RGAs序列均不同程度地含有NBS保守域的典型特征。Blast X分析发现,获得的4条魔芋RGAs与已知植物RGAs的氨基酸序列相似性为55%~76%。同源进化分析表明,这些魔芋RGAs均属于non-TIR-NBS-LRR类且可以分成3个亚类。本研究通过分子克隆技术获得花魔芋的抗病基因同源序列,为利用同源克隆技术克隆魔芋抗病基因作出探讨,为培育抗病新品种及抗病转基因育种提供参考。The isolation and analysis of the resistance gene analogs(RGAs)of konjac can provide reference for the cloning of the resistant genes of konjac.In this study,four pairs of degenerate primers were synthesized according to the conserved structural domains of the known plant disease-resistance genes,which amplified the disease-resistance genes of the flower konjac.Four resistant gene analogs were obtained.Putative amino acid sequences contained typical motifs of NBS conservative regions to some extent,and shared 20%~48%identities which know resistance genes in P-loop-GLPL region.Blast X analysis found that four KRGAs showed high sequence identities with known RGAs,and the amino acid identities were 55%~76%.Clustering analysis showed that the four RGAs were divided them into two subgroups and all belonged to non-TIR-NBS-LRR type of resistance genes.In this study,homologous sequences of disease-resistant genes of konjac were obtained by molecular cloning technology,which to explore the cloning of disease-resistant genes of konjac by homologous cloning technology,and provided a reference for breeding new disease-resistant varieties and disease-resistant transgenic breeding.
