运脾化浊解毒煎对泄泻型IBS大鼠SP、VIP、5-HT的影响

摘　　要：目的:探讨运脾化浊解毒煎对泄泻型肠易激综合征(IBS)大鼠P物质(SP)、血管活性肠肽(VIP)、5羟色胺(5-HT)的影响,为完善其药理机制提供数据支撑。方法:将Wistar大鼠随机分为6组:正常对照组、模型对照组、复方嗜酸乳杆菌0.05 g/kg组、运脾化浊解毒煎1.25 g/kg、2.5 g/kg、5.0 g/kg组。除正常对照组外,均以综合刺激法制备泄泻型IBS模型。模型制备成功后,各组分别给予相应药物或蒸馏水灌胃,连续2周。定时测定体重、摄食量、粪便含水量和腹壁撤退反射评分,测定肠推进率和肠粘膜形态学评分,分别采用ELISA法和免疫组织化学法测定血清及肠组织SP、VIP、5-HT水平。结果:与正常对照组相比,模型对照组肠推进率、形态学评分明显升高;血清和肠组织SP与5-HT水平明显升高,VIP水平明显降低。与模型对照组相比,复方嗜酸乳杆菌0.05 g/kg组和运脾化浊解毒煎各组肠推进率、形态学评分明显降低;血清和肠组织SP与5-HT含量或表达明显降低,VIP含量或表达明显升高。运脾化浊解毒煎1.25 g/kg组仅形态学评分降低,血清VIP水平升高。结论:运脾化浊解毒煎对改善泄泻型IBS大鼠脑-肠轴功能有积极作用,可能是其治疗IBS的重要机制之一。Objective:To explore the influence of Yunpi-Huazhuo-Jiedu decoction on SP,VIP and 5 HT of diarrhea IBS(Irritable Bowel Syndrome)in rats,so as to provide data base for its pharmacological mechanism.Methods:60 Wistar rats were divided into 6 groups:the control group,the model group,0.05 g/kg lactobacillus acidophilus group,Yunpi-Huazhuo-Jiedu decoction(1.25 g/kg、2.5 g/kg、5.0 g/kg).Except for the control group,all other rats were established models for cathartic IBS by comprehensive stimulation method.After modeling,rats were administered with corresponding drugs or saline,for constantly 2 weeks.The body weight,the food intake,the fecal water content and the abdominal wall withdrawal reflex score were determined at regular intervals,the intestinal propulsion rate and the intestinal mucosal morphological score were determined.the serum and tissue levels of SP,VIP and 5-HT were determined by ELISA and immunohistochemistry respectively.Results:Compared with the control group,the intestinal propulsion rate and the morphological score were significantly increased in the model group,serum and tissue levels of SP and 5-HT in the model group were significantly increased,the VIP level in the model group was significantly decreased.Compared with the model group,the intestinal propulsion rate and the morphological score in 0.05 g/kg lactobacillus acidophilus group and all Yunpi-Huazhuo-Jiedu decoction groups were significantly decreased,the serum and tissue levels of SP and 5-HT were significantly decreased,the serum and tissue levels of VIP were significantly increased.In 1.25 g/kg Yunpi-Huazhuo-Jiedu decoction group,the morphological score was decreased and serum VIP level was increased.Conclusion:Yunpi-Huazhuo-Jiedu decoction has the positive effect on improving brain-intestinal axis function of IBS with diarrhea in rats,which may be one of the important mechanisms to treat IBS.
