摘 要:长非编码RNA(long non?coding RNA,lncRNA)是一类长度超过200 nt并且缺乏蛋白质编码潜能的RNA分子。最初lncRNA被认为是由RNA聚合酶Ⅱ转录的副产物,且无生物学功能。随着转录组测序技术的发展,大规模的lncRNA被鉴定出来。越来越多的证据表明,lncRNA参与多种生物学过程,包括基因印记、基因组重排、染色质修饰、细胞周期调控、转录、剪接、mRNA降解和翻译。lncRNA异常表达与人类多种疾病相关,尤其是增生性疾病,包括胃癌、肝癌和直肠癌等。其中,睾丸相关的高度保守的致癌长非编码RNA(testis?associated highly?conserved oncogenic long non?coding RNA,THOR)是一种非常保守的非编码RNA,在睾丸中特异性表达,并广泛存在于人的多种肿瘤组织中,如肝癌、胃癌、鼻咽癌、肾细胞癌、骨肉瘤、视网膜母细胞瘤、黑色素瘤、非小细胞肺癌和舌鳞状细胞癌中,在其发生和发展过程中发挥重要作用。在鼻咽癌、肾细胞癌、骨肉瘤、黑色素瘤、非小细胞肺癌和舌鳞状细胞癌中,THOR主要通过与胰岛素样生长因子2 mRNA结合蛋白1(insulin?like growth factor 2 mRNA?binding protein 1,IGF2BP1)相互作用,促进肿瘤细胞的增殖。在肝癌中,THOR分别通过PTEN在胃癌和骨肉瘤中,THOR主要通过提高SOX9的表达增强癌细胞的干性。在视网膜母细胞瘤中,THOR主要通过提高c?myc的表达促进癌细胞增殖。在鼻咽癌中,THOR主要通过提高YAP的表达增强癌细胞的干性。Long non-coding RNA(lncRNA)is a class of RNA molecules that are more than 200 nt in length and lack protein-coding potential.Initially IncRNAs were thought to be a byproduct of RNA polymerase II transcription and had no biological function.With the development of the transcriptome sequencing technology,large-scale IncRNAs were identified.Increasing evidence suggest that IncRNAs are involved in a variety of biological processes including gene imprinting,genomic rearrangement,chromatin modification,cell cycle regulation,transcription,splicing,mRNA degradation and translation.Abnormal expression of lncRNA was associated with a variety of human diseases,especially proliferative diseases,including gastric cancer,liver cancer and rectal cancer.The testis-associated highly-conserved oncogenic long non-coding RNA(TH OR)is a highly conserved non-coding RNA that is specifically expressed in the testis and is widely present in various tumor tissues of humans,such as liver cancer,gastric cancer,nasopharyngeal carcinoma,renal cell carcinoma,osteosarcoma,retinoblastoma,melanoma,non-small cell lung cancer and tongue squamous cell carcinoma,which played an important role in carcinogenesis and development of tumors.In nasopharyngeal carcinoma,renal cell carcinoma,osteosarcoma,melanoma,non-small cell lung cancer and tongue squamous cell carcinoma,THOR could promote cell proliferation through binding to insulin-like growth factor 2 mRNA-binding protein 1(IG F2B P 1).In hepatocellular carcinoma,THOR could promote cell proliferation and liver cancer stem cells expansion through PTEN/AKT and p-catenin signaling,respectively.In gastric cancer and osteosarcoma,THOR could enhance the sternness of cancer cells by increasing the expression of S0X 9.In retinoblastoma,THOR could promote cell proliferation by increasing the expression of c-myc.In nasopharyngeal carcinoma,THOR could enhance the sternness of cancer cells by increasing the expression of YAP.
