摘 要:为了探究山药块茎膨大期淀粉合成、积累与淀粉合成关键酶及其相关调控基因表达的关系,以淀粉含量差异显著的河北安平白山药(Dioscorea opposita Thunb.)和毕克齐长山药(Dioscorea opposita Thunb.)为试验材料,测定块茎膨大期块茎中淀粉、支链淀粉、直链淀粉含量及比例(支链淀粉/直链淀粉)和淀粉合成关键酶(AMY、ADPGase、SPS、SSS)活性,并采用实时荧光定量PCR对块茎中淀粉合成关键酶基因(SPS 1、Isoamylase 3、PFK 3、Starch synthase 3)表达进行分析。结果表明,AMY、ADPGase及SSS对淀粉的合成起关键作用,ADPGase可能是直链淀粉合成的关键酶,SPS和AMY可能对支链淀粉合成起作用。PFK 3基因在山药淀粉合成过程中起重要作用,且PFK 3基因和Starch synthase 3基因可能对直链淀粉的合成起重要作用。In order to investigate the relationship between starch synthesis and accumulation and the expression of key enzymes and related regulatory genes in tuber expansion of yam,Hebei Anpin Bai yam(Dioscorea opposita Thunb.)and Bikeqi Chang yam(Dioscorea opposita Thunb.)with significant differences in starch content were used as test materials to determine the content and proportion of starch,amylopectin and amylose in tuber during tuber expansion(amylopectin/amylose)and the activity of key enzymes(AMY,ADPGase,SPS,SSS)in starch synthesis.The expression of key enzyme genes(SPS1,Isoamylase3,PFK3,Starch synthase3)of starch synthesis in tubers was analyzed by real-time fluorescence quantitative PCR.The results showed that AMY,ADPGase and SSS played a key role in starch synthesis,suggesting that ADPGase might be the key enzyme of amylose synthesis,and SPS and AMY might play an important role in amylopectin synthesis.PFK3 gene played an important role in starch synthesis of yam,and PFK3 gene and Starch synthase3 gene might play an important role in the synthesis of amylose.
