摘　　要：目的对血浆miR-499和miR-22作为急性心肌梗死(AMI)诊断标志物进行分析。方法本文纳入50例AMI患者,50例非心梗(N-MI)人群作为研究对象,采用实时荧光定量聚合酶链反应(qRT-PCR)分析miR-499和miR-22的表达水平。通过受试者操作特征(ROC)曲线分析临床诊断AMI的灵敏度和特异性。结果荧光定量PCR分析显示,miR-499的相对表达水平在HC、UAP和AMI三组间之间逐渐增加(P<0.05)。miR-22的表达则呈下降趋势(P<0.05)。Logistic回归显示miR-499和miR-22与AMI独立相关。ROC曲线显示当miR-499和miR-22水平分别为0.377和0.946时,诊断AMI的灵敏度分别为86.00%和86.00%,特异性分别为98.00%和94.00%。结论血浆miR-499和miR-22对AMI有一定的诊断效能,提示其可作为临床AMI诊断的潜在标志物。Objective To evaluate plasma miR-499 and miR-22 as diagnostic biomarkers in acute myocardial infarction.Methods In this study,50 patients with AMI(AMI)and 50 people with non-MI(N-MI)were included.The expression levels of plasma miR-499 and miR-22 were measured by real-time fluorescent quantitative polymerase chain reaction(qRT-PCR).The sensitivity and specificity of clinically diagnosed AMI were evaluated by receiver operating characteristic(ROC)curves.Results Real-time PCR results showed that the relative expression levels of miR-499 were gradually increased among the three groups(P<0.05),while the expression of miR-22 showed a downward trend(P<0.05).Logistic analysis showed levels of miR-499 and miR-22 were correlated to AMI independently.When the miR-499 and miR-22 levels were 0.377 and 0.946,the diagnostic values of miR-499 and miR-22 for AMI showed as following:86.00% and 86.00% for sensitivity,98.00% and 94.00% for specificity,respectively.Conclusion The results from the current study showed that plasma miR-499 and miR-22 were more sensitive and specific for the diagnosis of AMI,suggesting that they can be used as potential biomarkers for clinical diagnosis of AMI.
