摘 要:为了确定宿主的遗传背景对回归热赫姆斯疏螺旋体感染的影响,本研究选用了3种常用的不同遗传背景小鼠(C57BL/6、Balb/c和ICR),经腹腔注射感染回归热赫姆斯疏螺旋体,每天计数细菌和记录神经症状,并通过ELISA法分析血液中螺旋体特异性IgG及IgM抗体水平。结果表明,在整个21 d实验观察期内,回归热螺旋体在三种不同遗传背景小鼠体内均能高效复制,且菌血症呈周期性地发生。螺旋体特异性IgG抗体,随着感染时间推移而逐渐升高,并一直维持在较高的水平。螺旋体特异性IgM抗体呈现周期性地下降和回升。C57BL/6和ICR小鼠从感染后第9 d开始出现神经症状,并于第15 d达到高峰,然而ICR小鼠症状没有C57BL/6小鼠的严重。Balb/c小鼠在整个实验过程中没有出现明显的神经症状。综上所述,本研究表明宿主的遗传背景影响回归热螺旋体感染引起中枢神经性疾病的致病性,并且进一步确认了C57BL/6小鼠是所研究的3种小鼠中最为合适的回归热螺旋体感染引起中枢神经系统疾病模型。In order to determine effects of host’s genetic backgrounds on infection of relapsing fever(RF)Borrelia hermsii,three commonly used mouse strains with different genetic backgrounds(C57 BL/6,Balb/c and ICR)were intraperitoneally injected with the bacteria.Blood samples were collected from tail veins for bacterial counting,antibody detection via ELISA and neurological signs were recorded every day.The results showed that the RF Borrelia replicated efficiently in all 3 strains of mice.The bacteremia occurred periodically.The Borrelia-specific IgG in blood gradually increased and maintained at a high level.The Borrelia-specific IgM occurred periodically as bacteremia did.C57 BL/6 and ICR mice began to develop neurological signs at day 9 post infection and peaked at day 15.The neurological signs in ICR mice were less severe than those of C57 BL/6 mice.In contrast,Balb/c mice did not show obvious neurological signs during the whole experiment.In conclusion,the genetic background of hosts influenced the pathogenesis of relapsing fever Borrelia-induced central nervous system(CNS)diseases.Among three mouse strains used in this study,C57 BL/6 mice were confirmed to be the ideal experimental animals for further exploration of the CNS diseases caused by relapsing fever Borrelia hermsii.
