摘  要:宫川棘口吸虫是家禽常的寄生虫,偶尔也感染人。为了研究宫川棘口吸虫的分子进化地位,应用PCR方法扩增宫川棘口吸虫线粒体rrnS和rrnL序列,并以两个串联序列为标记基因,采取最大简约法(MP)构建系统发生树,探讨宫川棘口吸虫与其它吸虫的进化关系。结果扩增获的宫川棘口吸虫rrnS和rrnL序列序列长度分别为754 bp、992 bp;进化分析显示,除棘科吸虫外,其它虫体每科均形成一个独立分支。在棘口科吸虫的分支中,宫川棘口吸虫与除圆圃棘口吸虫外的其它棘口属吸虫聚集在一起,与传统形态学分类结果一致。表明线粒体rrnS和rrnL序列为吸虫分子进化分析的良好标记基因。Echinostoma miyagawai was a common parasite of poultry that also occasionally infected humans. In order to study the phylogeny of Echinostoma miyagawai,the mitochondrial DNA rrnS and rrnL sequences of E. miyagawai were amplified by polymerase chain reaction(PCR),and phylogenetic relationships of Digenea trematodes were reconstructed using maximum parsimony(MP)based on rrnS and rrnL sequences. These results showed that the length of the complete rrn S and rrn L sequence was 754 bp and992 bp,respectively. Phylogenetic analysis showed that every families formed independent group except family Echinostomatidae. In family Echinostomatidae group,E. miyagawai clustered together with other Echinostoma worms except Echinostoma hortense,it was consistent with the traditional morphological classification. It suggested that both mitochondrial DNA rrnS and rrnL sequences were a reliable genetic marker for phylogenetic analysis in trematodes.
