摘 要:目的探讨长链非编码RNA TP73反义RNA 1(TP73-AS1)在乳腺癌组织中的表达及临床意义。方法利用GEPIA在线数据库(包括TCGA和GTEx RNA测序表达数据)和从基因表达汇编(GEO)数据库中下载的乳腺癌相关数据集,分析乳腺癌组织的TP73-AS1水平及与预后的关系,采用Kaplan-Meier Plotter数据库分析TP73-AS1水平与乳腺癌预后的关系。采用实时定量PCR检测本院2016年1月至2017年12月手术切除的87对癌组织和癌旁组织的TP73-AS1水平,进一步分析其水平与乳腺癌临床病理特征的关系。结果GEPIA在线分析结果显示TCGA数据库中乳腺癌组织(n=1085)的TP73-AS1水平低于正常组织(P<0.05)。GEO数据集GSE20685(n=327)、GSE20711(n=88)和GDS3853(n=14)中乳腺癌组织的TP73-AS1水平分别为6.541±1.084、4.439±0.291和6.926±0.454,均低于正常乳腺组织的8.555±2.024(P<0.05)。GEPIA在线分析结果显示TCGA数据库中TP73-AS1表达与乳腺癌的总生存期(OS)和无病生存期(RFS)均无关(P>0.05),而Kaplan-Meier plotter在线分析显示TP73-AS1表达与乳腺癌的OS和RFS有关,而与无远处转移生存期和进展后生存期无关,其中高表达者的中位OS和RFS分别为110.4和48.0个月,高于低表达者的69.73和27.96个月(HR=0.71,95%CI:0.52~0.97;HR=0.62,95%CI:0.53~0.73)。87例乳腺癌组织的TP73-AS1水平为0.483±0.301,低于癌旁组织的1.475±0.704(P<0.05)。TP73-AS1水平与肿瘤大小、淋巴结转移、TNM分期、Ki-67表达和Nottingham预后指数有关(P<0.05),而与年龄、绝经、组织学分级、ER表达、PR表达和HER-2表达均无关(P>0.05)。结论TP73-AS1在乳腺癌中低表达且与肿瘤的发生发展和预后有一定关系,可用作筛查乳腺癌和预测预后的潜在标志物。Objective To investigate the expression of long non-coding RNA TP73 antisense RNA 1(TP73-AS1)in breast cancer tissues and its clinical significance.Methods Using the GEPIA online database(including TCGA and GTEX RNA sequencing expression data)and the breast cancer related data set downloaded from the gene expression omnibus(GEO)database,we analyzed the TP73-AS1 level in breast cancer tissues and its relationship with prognosis.Kaplan-Meier plotter database was used to analyze the relationship between TP73-AS1 level and prognosis of breast cancer.Real-time quantitative PCR was used to detect the TP73-AS1 level in 87 pairs of cancer tissues and paracancerous tissues surgically removed from January 2016 to December 2017 in our hospital.The relationship between the TP73-AS1 level and the clinicopathological characteristics of breast cancer was further analyzed.Results The results of GEPIA online analysis showed that the TP73-AS1 level of breast cancer tissue(n=1085)in TCGA database was lower than that of normal tissue(P<0.05).The TP73-AS1 levels of breast cancer tissues in GEO data sets GSE20685(n=327),GSE20711(n=88)and GDS3853(n=14)were 6.541±1.084,4.439±0.291 and 6.926±0.454,lower than 8.555±2.024 of normal breast tissues(P<0.05).The results of GEPIA online analysis showed that TP73-AS1 level was not related to overall survival(OS)and relapse-free survival(RFS),while Kaplan-Meier plotter online analysis showed that TP73-AS1 level was related to OS and RFS,but not to distant metastasis-free survival and progression-free survival.The median OS was 110.4 months in high-expression group,longer than 69.73 months in low-expression group(HR=0.71,95%CI:0.52-0.97).The median RFS was 48.0 months in high-expression group,longer than 27.96 months in low-expression group(HR=0.62,95%CI:0.53-0.73).The TP73-AS1 level was 0.483±0.301 in 87 cases of breast cancer tissues,lower than 1.475±0.704 in paracahcerous tissues(P<0.05).TP73-AS1 level was related with tumor size,lymph node metastasis,TNM stage,Ki-67 expression and Nottin
