摘  要:目的探讨血小板衍生生长因子受体(PDGFR)信号通路对血小板衍生生长因子BB(PDGF-BB)诱导的体外小鼠脑星形胶质细胞增殖的影响。方法取出生1~3 d的小鼠乳鼠大脑皮质培养星形胶质细胞,采用免疫荧光染色检测胶质纤维酸性蛋白(GFAP)的表达,对培养的星形胶质细胞的纯度进行鉴定。通过向培养细胞体系加入细胞因子PDGF-BB刺激细胞,应用PDGFR通路抑制剂AG1296进行干预,观察细胞增殖情况。Western blotting检测培养细胞系中p-PDGFR-α、p-PI3K、p-Akt及GFAP表达的变化。结果免疫荧光染色结果显示,星形胶质细胞的纯度可以达到90%。加入PDGF-BB后星形胶质细胞增殖明显增多,给予抑制剂AG1296预处理增殖受到抑制。Western blotting结果显示,加入PDGF-BB后p-PDGFRα、p-PI3K、p-Akt及GFAP表达增多(P<0.01),给予抑制剂预处理表达减少(P<0.05)。结论PDGFR信号通路参与体外小鼠脑星形胶质细胞的增殖。Objective To investigate the effects of the platelet-derived growth factor receptor(PDGFR)signaling pathway on the proliferation of mouse brain astrocytes induced by platelet-derived growth factor-BB(PDGF-BB).Methods Astrocytes were isolated from the cerebral cortex of mice from day 1 to day 3 after birth.The purity of cultured astrocytes was identified by immunofluorescence staining of glia fibrillary acidic protein(GFAP).Cultured cells were stimulated with PDGF-BB and treated with a PDGFR inhibitor,AG1296.Cell proliferation was then observed.The expression of p-PDGFR-α,p-PI3Kt,p-Akt,and GFAP was detected by Western blotting.Results The purification and identification of astrocytes showed that astrocyte cell purity reached 90%.Astrocyte proliferation increased significantly with the addition of PDGF-BB and decreased upon pre-treatment with AG1296.Further,Western blotting showed that the expression of p-PDGFR-α,p-PI3Kt,p-Akt,and p-GFAP increased after the addition of PDGF-BB(P<0.05)and decreased after pre-treatment with AG1296(P<0.05).Conclusion The PDGFR signaling pathway promotes the proliferation of mouse brain astrocytes in vitro.
