摘  要:目的观察鹿茸多肽对冠脉结扎心肌缺血模型大鼠心肌特异性转录因子表达的影响。方法Wistar大鼠随机分为6组,心脏左冠状动脉前降支结扎复制大鼠心肌缺血损伤模型,连续给药28d。PowerLab数据采集分析系统检测各组大鼠心率及心电图ST段变化,Elisa法检测各组大鼠血清中Nkx2.5、GATA4、ATF-2和MEF-2C含量,苏木精-伊红(HE)染色观察大鼠心肌组织病理学形态变化。结果鹿茸多肽可明显降低心脏左冠状动脉前降支结扎所致心肌缺血损伤大鼠模型的心率,ST段上调幅度明显降低,升高大鼠血清中Nkx 2.5、GATA4和MEF-2C含量(P<0.05),ATF-2无明显变化(P>0.05)。结论鹿茸多肽可明显缓解冠状动脉结扎所致的大鼠心肌缺血损伤,其作用机制可能与调节心肌特异性转录因子Nkx 2.5、GATA4、MEF-2C水平有关。Objective:To observe the effect of velvet antler polypeptide on the expression of myocardial specific transcription factors Nkx 2.5,GATA4,ATF-2,MEF-2C in coronary artery ligation myocardial ischemia model rats.Methods:Randomly grouped,coronary ligation was used to duplicate myocardial ischemia model,and the drug was administered for 28 days.Heart rate and electrocardiogram of rats in each group were detected,Nkx2.5,GATA4,ATF-2 and MEF-2C contents in serum of rats in each group were detected by Elisa method,and pathological changes of myocardial tissue were observed by HE staining.Results:Velvet antler polypeptide can obviously change the heart rate and electrocardiogram of rats with myocardial injury caused by coronary artery ligation,and increase the contents of Nkx 2.5,GATA4 and MEF-2C in rat serum(P<0.05),while ATF-2 has no obvious change(P>0.05).Conclusion:Velvet antler polypeptide can significantly alleviate myocardial injury induced by ligation of the coronary artery in rats,and its mechanism may be related to the regulation of myocardial specific transcription factors Nkx2.5,GATA4 and MEF-2C.
