摘 要:通过对泡泡叶杜鹃(Rhododendron edgeworthii Hook. f.)实生幼苗的组织培养,探索适宜泡泡叶杜鹃的灭菌方法及增殖培养基配方,以建立泡泡叶杜鹃的组织培养体系。结果表明,用0.1%的氯化汞灭菌8 min泡泡叶杜鹃污染率最低,为7.5%;当培养基中噻苯隆(TDZ)浓度为0.3 mg/L、吲哚丁酸(IBA)浓度为0.5 mg/L时,泡泡叶杜鹃的芽诱导系数最高,达到5.20;当培养基中TDZ浓度为0.5 mg/L、IBA浓度为0.05 mg/L时,芽分化系数最高,达到13.00;当培养基中激动素(KT)浓度为0.8 mg/L、IBA浓度为0.08 mg/L、赤霉素(GA3)浓度为0.2 mg/L时,泡泡叶杜鹃的壮苗率最高,达到96.00%。Through the tissue culture of Rhododendron edgeworthii Hook. f. seedlings, the sterilization method and proliferation culture formula of it were explored to establish the tissue culture system. The results shows that, the contamination rate of Rhododendron edgeworthii Hook. f. was 7.5% when they were sterilized in 8 min treatment under 0.1% HgCl2;the bud induction coefficient of Rhododendron edgeworthii Hook. f. was the highest, reaching 5.20, when the concentration of TDZ was 0.3 mg/L and the concentration of IBA was 0.5 mg/L;the bud differentiation coefficient of Rhododendron edgeworthii Hook. f. was the highest, reaching13.00, when the concentration of TDZ was 0.5 mg/L and the concentration of IBA was 0.05 mg/L;when the concentration of KT was0.8 mg/L, the concentration of IBA was 0.08 mg/L, and the concentration of GA3 was 0.2 mg/L, the growth rate of the rhododendrons was the highest, reaching 96.00%.
