摘 要:目的:探讨产后抑郁(postpartum depression,PPD)模型大鼠前额叶皮质、海马、下丘脑雌激素受体(estrogen receptor,ER)表达分布和变化规律以及电针对ER的影响。方法:选用雌性5~6月龄SD大鼠,随机分为正常组、假手术组、模型组、电针组、西药组,每组均设2、4、6周观察时点。采用免疫组化法测定大鼠前额叶皮质、海马、下丘脑ERα与ERβ阳性细胞表达数。结果:模型组大鼠第2、4、6周前额皮质、海马、下丘脑ERα表达较正常组、假手术组减少,差异有统计学意义(P<0.05);电针组、西药组第2、4、6周前额皮质、海马、下丘脑ERα表达均较模型组增加,差异有统计学意义(P<0.05);模型组第2周前额皮质,第4、6周前额皮质、海马、下丘脑ERβ表达较正常组、假手术组减少,差异有统计学意义(P<0.05),第2周海马、下丘脑ERβ表达较正常组、假手术组增加,差异有统计学意义(P<0.05);电针组第6周前额叶皮质,第2、4、6周海马,第4、6周下丘脑,西药组第4、6周前额叶皮质,第2、4、6周海马,第4、6周下丘脑ERβ表达较模型组增加,差异有统计学意义(P<0.05)。结论:前额叶皮质、海马、下丘脑ERα与ERβ表达水平的改变可能是PPD模型大鼠PPD发生发展的生物学基础之一,电针对产后抑郁模型大鼠脑组织内ERα和ERβ表达量的调节可能是电针治疗PPD的重要作用机制之一。Objective:To explore the distribution law and expressions of estrogen receptor(ER)of brain cortex,hippocampus and hypothalamus in the rats suffering postpartum depression(PPD),and the effects of acusector on ER.Methods:SD female rats,aged between five and six months,were selected and randomly divided into the normal group,sham operation group,the model group,acusector group and Western medicine group.All the groups were observed at the second,forth and sixth week.Immunohistochemical method was used to detect the expressions of positive cells ERαand ERβin prefrontal cortex,hippocampus and hypothalamus.Results:The expressions of ERαin prefrontal cortex,hippocampus and hypothalamus of the model group lowered compared with the normal group and sham operation group at the second,forth and sixth week,and the difference could be found(P<0.05).The expressions of ERαin prefrontal cortex,hippocampus and hypothalamus of acusector group and Western medicine group at the second,forth and sixth week increased compared with the model group,and the difference could be found(P<0.05).The expressions of ERβin prefrontal cortex at the second week,prefrontal cortex,hippocampus and hypothalamus at the forth week and sixth week of the model group reduced compared with the normal group and sham operation group,and the difference showed statistical meaning(P<0.05),the expressions of ERβin hippocampus and hypothalamus increased at the second week compared with the normal group and sham operation group,and the difference showed statistical meaning(P<0.05).The expressions of hypothalamus at the forth and sixth week,prefrontal cortex at the forth and sixth week,hippocampus at the second,forth and sixth week,hypothalamus at the forth and sixth week of the Western medicine group increased compared with the model group,and the differences were statistically significant(P<0.05).Conclusion:The expression changes of ERαand ERβin prefrontal cortex,hippocampus and hypothalamus might be one factor of biological basis of the incidence and deve
