摘  要:以寄生植物桑寄生种子为研究对象,研究2,3,5-氯化三苯基四氮唑(TTC)的浓度、染色时间、染色温度对桑寄生种子生活力测定的效果;分析桑寄生种子生活力与萌发率的相关性,探究桑寄生种子顽拗性生物学特征以及保存方法。结果表明,TTC法能快速测定桑寄生种子的生活力,其染色最佳条件为30℃条件下0.8%TTC溶液里浸种8 h。桑寄生种子生活力与萌发率的相关系数为0.990,并得到线性回归方程Y=0.936 X-8.956。桑寄生种子的含水量临界值约为30%,低温保存临界值为0℃。桑寄生种子在2 mg·L^-1的ABA溶液浸种30 min,在4℃的冰箱保存可有效延缓种子生活力丧失。In order to investigate the effects of the concentration,staining time and temperature of 2,3,5-triphenyl tetrazolium chloride(TTC) on the viability of Taxillus chinensis(DC.) Danser seeds,the correlation between viability and germination rate was analyzed,and the biologic characteristics of recalcitrance and conservation technology of T. chinensis seeds were also studied.The results showed that TTC could be used to determine the viability of the seeds rapidly,and the optimal dyeing condition was to immerse the seeds in 0.8% TTC solution at 30 ℃ for 8 h.The correlation coefficient between viability and germination rate was 0.990,and the linear regression equation Y=0.936 X-8.956 was obtained.The critical value of water content in T. chinensis seeds was about 30% and the critical value of low temperature preservation was 0 ℃.Additionally,the seeds were immersed in 2 mg·L^-1 ABA solution for 30 min and stored at 4 ℃ in a refrigerator,which could effectively delay the loss of seed viability.
