摘 要:目的:为了筛选新颖有效的乙酰胆碱酯酶(AchE)和单胺氧化酶(MAO)抑制剂,对延胡索总生物碱进行了提取和分离。方法采用大孔吸附树脂法,硅胶色谱法和TLC法研究其化学成分并运用酶标法测定单体化合物乙酰胆碱酯酶和单氨氧化酶抑制活性。结果从延胡索中分离8个化合物其中化合物4和化合物8是AchE新的可逆的非竞争性抑制剂,它们的半数抑制浓度分别为9.27μM和20.98μM,化合物7是新的选择性单氨氧化酶B(MAO-B)抑制剂JC50值为50.94μM。结论:延胡索中存在AchE和MAO抑制剂。Objective:To screen novel and effective acetylcholinest erase(AchE) and monoaminoxidase(MAO)inhibitors by drawing and separating total al kal oi ds(TA) from Yanhusuo(Corydalis yanhusuo WT.Wang).Methods:Macroporous resin adsorption was adopted,silicagel chromatography and TLC method were used to study chemical ingredi ents,ELISA was used to measure the inhi bitory activity of AchE and Results:Eight compounds were separated from Yanhusuo,among them,4 and 8 were new,reversi bl e noncompeti ti ve i nhi bi t ors of AchE,IC50 were 9.27μM and 20.98 μM,7 was a new sel ective MAQ Binhibitor,IC50 was 50.94 μM Conclusion:AchE and MAO inhibitors are existed in Yanhusuo.
