孔雀五彩芋的离体再生与繁殖

摘  要:以成熟叶片为外植体,建立五彩芋离体再生技术,观察再生植株移栽后的生长表现.结果表明,在含2 mg/L BA和2 mg/L NAA的MS培养基上培养60 d,60%的外植体可以形成愈伤组织;在含2或3 mg/L BA和0.2 mg/L NAA培养基上,愈伤组织可以100%地分化不定芽,不定芽继代在2 mg/L BA和0.2 mg/L NAA培养基上,50 d可以增殖6倍以上;在含1或2 mg/L IBA培养基上,所有的芽均可在一个月内生根、形成完整的再生植株.再生植株移栽后成活率高,生长正常,移栽3个月后所形成的所有叶片出现彩斑,彩斑的类型与母株性状一致.The regeneration of Makoyana(Caladium hortulanum)in vitro was established using mature leaves as explants,and then the growth performance of the regenerated plants was examined.The results showed that 60%of the explants formed callus within 60 days of culture on MS medium containing 2 mg/L BA and 2 mg/L NAA,all callus pieces initiated adventitious buds when they were incubated on the media supplemented with 2 or 3 mg/L BA plus 0.2 mg/L NAA for 60 days,and the buds propagated very well when they were subcultured on the medium with 2 mg/L BA and 0.2 mg/L NAA and also all the buds generated root to grow the intact plants within one month on 1 or 2 mg/L IBA medium.The regenerated plants survived very well and grew normally after they were cultivated in greenhouse,and all leaves formed after 3 months of cultivation and showed the variegation pattern identical to the mother plants.
