摘 要:目的探讨砷对雄性大鼠精子生成及血清睾酮浓度的影响。方法40只雄性SD大鼠随机分成5组,分别是空白对照组(生理盐水)、阳性对照组(100μg·kg^-1雌二醇)和0.15、0.75、1.50 mg·kg^-1(低、中、高剂量)砷染毒组,每组8只。每天灌胃1次,连续染毒8周后,观察大鼠睾丸、附睾组织病理学改变并对其精子数量和存活率、血清睾酮水平进行测定。结果病理学结果显示:空白组大鼠睾丸组织曲细精管结构完整、排列紧密,生精细胞层次清楚;阳性对照组和各砷染毒组曲细精管萎缩、管腔间隙变大,生精细胞排列紊乱。空白组大鼠附睾管结构完整,内含大量精子;阳性对照组和各砷染毒组大鼠附睾管腔间隙变大,精子数减少。各砷染毒组大鼠的精子数量、精子存活率均低于空白对照组(P均<0.05);低砷染毒组大鼠的精子数量高于阳性对照组(P<0.05),低、中剂量砷染毒组大鼠的精子存活率均高于阳性对照组(P均<0.05)。高剂量砷染毒组大鼠血清睾酮浓度均低于空白对照组和阳性对照组(P均<0.05)。结论砷对雄性大鼠精子的生成具有抑制作用,其抑制效果与雌二醇相当,砷还可使血清睾酮浓度下降。Objective To investigate the effects of arsenic on sperm production and serum testosterone concentration in male rats.Methods Forty male Sprague-Dawley rats were randomly divided into 5 groups,with 8 rats in each group.They were blank control group(normal saline),positive control group(100μg·kg^-1 estradiol)and 0.15 mg·kg^-1,0.75 mg·kg^-1,1.5 mg·kg^-1 low,medium and high doses of arsenic exposure groups.After exposure for 8 weeks,the pathological changes of testis and epididymis were observed.The number and survival rate of the concentration of serum testosterone were measured.Results Pathological results showed that the structures of the seminiferous tubules of the rats in the blank group were complete and compact,spermatogenic cells were well organized.In the positive control group and arsenic-exposed groups,the seminiferous tubules shrinked,the lumen clearance became larger,the spermatogenic cells were disorganized.The rats in the blank group had complete epididymal duct structure and contained a large number of sperm.In the positive control group and arsenic-exposed groups,the epididymal tubules lumen clearance became larger,the sperm count decreased.The number and survival rate of sperm in each arsenic poisoning group were lower than those in the blank control group(P all<0.05).The sperm count of the low arsenic poisoning group was higher than that of the positive control group(P<0.05).The sperm survival rate of the low and medium dose arsenic poisoning groups were higher than that of the positive control group(P all<0.05).The concentration of serum testosterone in the high-dose arsenic poisoning group was lower than that in the blank control group and the positive control group(P all<0.05).Conclusion Arsenic has inhibitory effect on sperm production in rats which is similar to estradiol.Arsenic can also reduce the concentration of serum testosterone.
