摘 要:目的观察生精逐瘀汤对精索静脉曲张(VC)大鼠睾丸低氧诱导因子1α(HIF-1α)、p53 mRNA及蛋白表达的影响。方法取6只SD雄性大鼠作为假手术组(仅显露左肾静脉,但不结扎)。其余大鼠进行左肾静脉结扎手术造模后,适应性喂养1周,将造模成功的24只大鼠随机分为模型组及生精逐瘀汤低、中、高剂量组,每组6只。造模组予0.9%氯化钠注射液2 mL/只灌胃,生精逐瘀汤低、中、高剂量组分别予15、30、60 g/kg生精逐瘀汤灌胃,每日1次,共12周。假手术组未予任何处理。于造模术后13周处死大鼠,比较体质量、睾丸指数及附睾指数,左侧精索静脉直径,检测各组大鼠睾丸组织的HIF-1α、脯氨酸羟化酶2(PHD2)、B淋巴细胞瘤-2相关x蛋白(Bax)、p53的mRNA及蛋白表达水平。结果与假手术组比较,生精逐瘀汤高剂量组大鼠体质量降低(P<0.05),模型组及生精逐瘀汤低剂量组大鼠睾丸指数均降低(P<0.05);与模型组比较,生精逐瘀汤高剂量组大鼠体质量降低(P<0.05),生精逐瘀汤低、中、高剂量组大鼠睾丸指数均高于模型组(P<0.05)。与假手术组比较,模型组及生精逐瘀汤低、中、高剂量组大鼠左侧精索静脉直径均增加(P<0.05);与模型组比较,生精逐瘀汤低、中、高剂量组大鼠左侧精索静脉直径均增加(P<0.05)。与假手术组比较,生精逐瘀汤低剂量组HIF-1αmRNA表达水平上调(P<0.05),模型组Bax mRNA表达水平上调(P<0.05),模型组及生精逐瘀汤中、高剂量组p53 mRNA表达水平下调(P<0.05);与模型组比较,生精逐瘀汤低剂量组HIF-1αmRNA表达水平上调(P<0.05),生精逐瘀汤中、高剂量组Bax mRNA表达水平下调(P<0.05),生精逐瘀汤中剂量组p53 mRNA表达水平下调(P<0.05);与生精逐瘀汤低剂量组比较,生精逐瘀汤中、高剂量组HIF-1αmRNA表达水平下调(P<0.05),生精逐瘀汤中剂量组p53 mRNA表达水平下调(P<0.05);与生精逐瘀汤中剂量组比较,生精逐瘀汤高剂量组p53 mObjective To observe the effect of Shengjing Zhuyu decoction on hypoxia inducible factor-1α(HIF-1α),p53 mRNA and protein expression in testes of varicocele(VC) rats. Methods Six male SD rats were randomly selected as the sham operation group(only the left renal vein was exposed, but not ligated). The other rats underwent left renal vein ligation for modle and adaptive feeding for one week. 24 rats were randomly divided into the model group and the low, middle and high dose group of Shengjing Zhuyu decoction, with 6 rats in each group. The model group was given 0.9% sodium chloride injection of equal volume by gavage. The low, medium and high dose groups was given 15, 30 and 60 g/kg Shengjing Zhuyu decoction by gavage once a day for 12 weeks. No treatment was given to the sham operation group. At the 13 th week after operation, the rats were killed. The body mass, testis mass and epididymis mass, the dilated diameter of left spermatic vein were measured. The mRNA and protein expression of HIF-1α, proline hydroxylase-2(PHD2), b-lymphoma-2-related X protein(Bax) and p53 mRNA and protein expression were detected. Results Compared with the sham operation group, the body mass of the high dose group was decreased(P<0.05), the testis index of the model group and the low dose group was decreased(P<0.05);Compared with the model group, the body mass of the high dose group of Shengjing Zhuyu decoction was decreased(P<0.05), and the testis index of the low, middle and high dose group of Shengjing Zhuyu decoction was higher than that of the model group(P<0.05). Compared with sham operation group, the diameter of left spermatic vein in model group and low, middle and high dose group of Shengjing Zhuyu decoction was increased(P<0.05);Compared with model group, the diameter of left spermatic vein in low, middle and high dose group of Shengjing Zhuyu decoction was increased(P<0.05).
