摘 要:目的探讨槐耳清膏对人肾小管上皮细胞(HK-2细胞)内质网应激的保护作用。方法将HK-2细胞分为对照组(正常HK-2细胞),毒胡萝卜素(TG)组(5μmol/L TG处理8 h),5 mg/mL、10 mg/mL中药组(5、10 mg/mL槐耳清膏处理12 h),中药+TG组(5、10 mg/mL槐耳清膏处理4 h后加入5μmol/L TG共同作用8 h)。采用Western blotting检测各组细胞内质网源性转录因子(CHOP)和内质网分子伴侣葡萄糖调节蛋白78(GRP78)的表达情况。结果与对照组相比,TG组细胞CHOP和GRP78的表达显著增加(P<0.05),中药组CHOP及GRP78表达无明显变化(P>0.05)。与TG组相比,中药(5、10 mg/mL)+TG组CHOP与GRP78表达显著减少。结论TG可诱导人肾小管上皮HK-2细胞发生内质网应激反应。适宜浓度的槐耳清膏可能通过下调CHOP、GRP78表达,抑制HK-2细胞内质网应激反应,从而发挥保护肾小管上皮细胞的作用。Objective To investigate the protective effect of the Chinese herbal medicine Huaierqing on human renal tubular epithelial cells(HK-2 cells)with endoplasmic reticulum stress(ERS).Methods The cells were divided into the following six groups:control(normal HK-2 cells),thapsigargin(TG)(,HK-2 cells treated with 5μmol/L TG for 8 h),5 or 10 mg/mL Chinese herbal medicine(HK-2 cells treated with 5 or 10 mg/mL Chinese herbal medicine Huaierqing for 12 h),and 5 or 10 mg/mL Chinese herbal medicine+TG(HK-2 cells treated with 5 or 10 mg/mL Chinese herbal medicine Huaierqing for 4 h,followed by 5μmol/L TG treatment as a coefficient reaction for 8 h).Western blotting analysis was used to measure the expression of glucose-regulated protein(GRP)78 and C/-EBP homologous protein(CHOP).Results Compared with the control group,CHOP and GRP78 were markedly overexpressed in the TG group(P<0.05);however,they were not expressed significantly in both the Chinese herbal medicine groups(P>0.05).Compared with the TG group,CHOP and GRP78 expression levels were significantly downregulated in the 5 or 10 mg/mL Chinese herbal medicine+TG groups(P<0.05).Conclusion TG induced ERS in HK-2 cells.At the appropriate concentration,the Chinese herbal medicine Huaierqing exhibited a protective effect by reducing the expression of CHOP and GRP78,which inhibited ERS in HK-2 cells.
