摘 要:通过花特异启动子AP3和RNA酶Barnase基因构建了AP3-Barnase雄性不育基因表达盒,并获得了Columbia型转基因拟南芥。通过分子检测、GFP观察和育性分析,结果表明通过花特异启动子AP3调控不育基因Barnase可获得雄性不育植株。表型观察发现对所获得的转基因拟南芥除花器官与野生型对照有差异外,其他表性特征与野生型对照一致。主要差异表现为花瓣缺失、花丝缩短、花药位于柱头约1/3处、花粉活力下降或无活力、角果结实数量稀少甚至不能结实等表型特征。对后代种子的发芽试验结果表明利用AP3启动子驱动Barnase基因可获得雄性不育株,且不影响后代种子的正常发芽过程。该研究结果为雄性不育材料的创制提供了新的基因组合策略和功能参考。In this study,AP3-Barnase male sterility gene expression box was constructed with flower specific promoter AP3 and RNA enzyme gene Barnase,and transgenic Columbia Arabidopsis thaliana was obtained.By molecular detection,GFP observation and fertility analysis,the results showed that sterility gene Barnase could generate male sterile plants with the control of flower specific promoter AP3.The transgenic Arabidopsis have no differences with the wild type controls except the flower organs,and other phenotype characteristics were consistent with the wild type controls.The main differences include the lost petals,the shorter filaments,the anther dwarf and locating at about 1/3 stigma,the declined pollen vitality or no vigor,and the scarcity of fruit from pods or even unfruitful.The offspring seeds germination test results showed that male sterile strains could be obtained by using AP3 promoter to drive Barnase,and without affecting normal germination process of offspring seeds.These results provide new gene combination strategies and functional references for the development of male sterile materials.
