摘 要:为明确转录因子ZmMYC7与JAZ家族基因的互作关系,本研究构建了ZmMYC7与拟南芥JAZ家族基因的酵母双杂交载体,利用酵母双杂交技术研究ZmMYC7与拟南芥JAZ家族基因的互作关系。结果发现,共转化BD-ZmMYC7与AD-AtJAZ1、AD-AtJAZ2、AD-AtJAZ3、AD-AtJAZ4、AD-AtJAZ5、AD-AtJAZ8、AD-AtJAZ9、AD-AtJAZ10、AD-AtJAZ12组合的酵母在三缺培养基(SD/-T/-L/-H)和四缺培养基(SD/-T/-L/-H/-A)上均能正常生长,而BD-ZmMYC7与AD-AtJAZ6、AD-AtJAZ7、AD-AtJAZ11组合的酵母以及阴性对照组均不能在三缺、四缺培养基上正常生长,表明ZmMYC7与AtJAZ1、AtJAZ2、AtJAZ3、AtJAZ4、AtJAZ5、AtJAZ8、AtJAZ9、AtJAZ10、AtJAZ12能在酵母细胞中直接互作。研究结果为进一步研究转录因子ZmMYC7在JA信号防御反应中的功能及机制奠定了基础。In order to clarify the interaction between transcription factor ZmMYC7 and JAZ family genes,the yeast two-hybrid vectors of ZmMYC7 and Arabidopsis JAZ family genes were constructed in this study.The yeast twohybrid technology was used to study the interaction between ZmMYC7 and Arabidopsis JAZ family genes.Compared with the negative controls,the yeast colonies which were co-transformed with BD-ZmMYC7 plus AD-AtJAZ1,AD-AtJAZ2,AD-AtJAZ3,AD-AtJAZ4,AD-AtJAZ5,AD-AtJAZ8,AD-AtJAZ9,AD-AtJAZ10,and AD-AtJAZ12 vectors grew well in both SD/-T/-L/-H and SD/-T/-L/-H/-A media in yeast two-hybrid assay.However,the yeast colonies which were co-transformed with BD-ZmMYC7 plus AD-AtJAZ6,AD-AtJAZ7,AD-AtJAZ11 vectors and negative controls could not grow normally in both SD/-T/-L/-H and SD/-T/-L/-H/-A media.These results indicated that ZmMYC7 directly interacts with AtJAZ1,AtJAZ2,AtJAZ3,AtJAZ4,AtJAZ5,AtJAZ8,AtJAZ9,AtJAZ10 and AtJAZ12 in yeast cells.This study laid a foundation for further study on the function and mechanism of transcription factor ZmMYC7 in JA signal defense response.
