摘 要:转录因子MYB73在拟南芥抵抗生物和非生物胁迫过程中发挥重要作用,但其作用的分子机制尚待探究。本研究利用酵母双杂交(Yeast two-hybrid,Y2H)技术,对拟南芥MYB73与TPRs(TOPLESS-related proteins)蛋白之间的互作关系进行研究,结果发现,共转化MYB73与TPR3、TPR4组合(BD-MYB73+AD-TPR3、AD-MYB73+BD-TPR3、BD-MYB73+AD-TPR4、AD-MYB73+BD-TPR4)的酵母能够在三缺(SD/-L/-T/-H)和四缺(SD/-L/-T/-H/-A)培养基上生长,而共转化MYB73与TPR1、TPR2组合的酵母以及阴性对照组均不能在三缺、四缺培养基上正常生长,表明MYB73与TPR3、TPR4蛋白互作,与TPR1、TPR2不互作;与阳性对照相比,共转化EAR(Ethylene-responsive element binding factor-associated amphiphilic repression)基序突变的MYB73-m1(L197V)、MYB73-m2(L201V)与TPR3、TPR4组合(AD-MYB73-m1+BD-TPR3、ADMYB73-m2+BD-TPR3、AD-MYB73-m1+BD-TPR4、AD-MYB73-m2+BD-TPR4)的酵母不能在三缺和四缺培养基上生长,表明MYB73通过EAR基序与TPRs蛋白互作。Transcription factor MYB73 plays an important role in regulating Arabidopsis resistance to biotic and abiotic stress,but the molecular mechanism of its role remains to be explored.In this study,yeast two-hybrid technology was used to study the interaction between MYB73 and TPRs protein.The yeast colonies which were co-transformed with MYB73 plus TPR3 and TPR4(BD-MYB73+AD-TPR3,AD-MYB73+BD-TPR3,BDMYB73+AD-TPR4 and AD-MYB73+BD-TPR4)combinations grew well in three deficient media(SD/-L/-T/-H)and four deficient media(SD/-L/-T/-H/-A).However,the yeast colonies which were co-transformed with MYB73 plus TPR1,TPR2 combinations and negative controls could not grow normally in both three and four deficient media.These results indicated that MYB73 interacts with TPR3 and TPR4 proteins,but not with TPR1 and TPR2 in yeast cells.Compared with the positive controls,the yeast colonies which were co-transformed with EAR(Ethylene-responsive element binding factor-associated amphiphilic repression)motif mutation MYB73-m1(L197V),MYB73-m2(L201V)plus TPR3,and TPR4 combinations(AD-MYB73-m1+BD-TPR3,AD-MYB73-m2+BDTPR3,AD-MYB73-m1+BD-TPR4 and AD-MYB73-m2+BD-TPR4)could not grow on the three or four deficient media.These results indicated that MYB73 interacts with TPRs proteins through the EAR motif.
