摘  要:为研究枯草芽孢杆菌(Bacillus subtilis)YB1制备的秸秆生物炭固定化菌剂对烟嘧磺隆(Nicosulfuron)的降解效果,本试验通过单因素试验对生物炭固定YB1的条件进行优化和筛选。在此基础上,利用高效液相色谱检测固定化YB1菌剂在液体培养基中对烟嘧磺隆的降解情况,同时利用室内盆栽试验,以油菜为指示植物研究土壤中固定化YB1菌剂对烟嘧磺隆的降解效果。结果显示,培养基中YB1菌落数量为5.8×10^8cfu/mL,添加80~200目生物炭20 mg/mL,35℃固定24 h为最佳条件,生物炭上YB1的菌落数量约为5.5×10^9 cfu/g。高效液相色谱检测结果显示,固定化YB1菌剂对烟嘧磺隆具有较好的降解效果,在LB培养基中添加15 mg/mL固定化YB1菌剂,30℃培养72h,可降解92.51%的1.25mg/L烟嘧磺隆。室内盆栽试验结果表明,油菜株高和烟嘧磺隆浓度呈线性关系,在土壤中添加10 g/kg固定化YB1菌剂可以明显缓解烟嘧磺隆对油菜生长的抑制,抑制率由69.43%降至36.78%,表明生物炭固定化YB1菌剂对烟嘧磺隆有良好的降解效果。To study the degradation effect of biochar immobilized bacteria prepared by Bacillus subtilis YB1 on nicosulfuron,the conditions of biochar immobilized YB1 strain were optimized and screened by single factor test.Based on that,high performance liquid chromatography(HPLC)was used to detect the nicosulfuron degradation in liquid medium by immobilized YB1 bacteria,and at the same time,the pot experiment was used to study the nicosulfuron degradation in the soil by immobilized YB1 bacterium with rape as the indicator plant.The results showed that the best condition was the number of YB1 colony in the medium as 5.8×10^8 cfu/mL,adding 80-200 mesh biochar 20 mg/mL and immobilizing at 35℃ for 24 h.The colonies number of YB1 on biochar was approximately 5.5×10^9 cfu/g.The HPLC results showed that the immobilized YB1 bacterium had good degradation effect on nicosulfuron.Adding 15 mg/mL immobilized YB1 bacterium to LB medium could degrade 92.51%of nicosulfuron with the concentration of 1.25 mg/L after reacting at 30℃ for 72 h.The results of pot experiment showed that the rape height and the concentration of nicosulfuron had a linear relationship.Adding10 g/kg of immobilized YB1 to soil,the inhibition of nicosulfuron on rapeseed growth could be relieved obviously,and the inhibition rate was decreased from 69.43%to 36.78%.It indicated that the biochar immobilized YB1 bacterium has a good degradation effect on nicosulfuron.
